田基黄总黄酮抗CCl4复合因素所致大鼠肝纤维化的实验研究 被引量:3

Experimental Study of Therapeutic Effects of Total Flavonoids of Hypericum Japonicum Against Liver Fibrosis Induced by CCl4 Composited Factor in Rats

摘要 目的:研究田基黄总黄酮抗大鼠肝纤维化作用。方法:采用CCl4复合因素诱导肝纤维化大鼠模型;从第7周起,每天灌胃给予田基黄总黄酮(9、18、36mg/kg)1次,连续给药12周后,测定血清总胆红素(TBIL)、直接胆红素(DBIL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、TNF-α水平及肝组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)和羟脯氨酸(Hyp)的含量及MMP-1和TIMP-1蛋白的表达。结果:田基黄总黄酮能显著降低CCl4复合因素诱导的肝纤维化大鼠血清中TBIL、DBIL、ALT、AST、PC-Ⅲ、HA、LN的水平、肝组织中Hyp的含量和MMP-1和TIMP-1蛋白的表达,说明田基黄总黄酮具有良好的抗胆汁性肝纤维化的作用。此外,田基黄总黄酮也能提高肝组织SOD、GSH-Px的活性,降低肝组织MDA及血清中TNF-α的含量。结论:田基黄总黄酮可抑制CCl4复合因素诱导的大鼠肝纤维化的形成,其抗肝纤维化作用可能与其抗氧化作用及抗TNF-α的分泌有关。 Objective:The purpose of this study was to investigate the therapeutic effects of total flavonoids of Hypericum japonicum against liver fibrosis induced by CCl4 composited factor in rats. Methods: Liver fibrosis model was established by administration of CCl4 composited factor daily for 6 weeks. From the 7th week, the treatment groups orally received total flavonoids of Hypericum japonicum(9、18、36mg/kg/day) daily for 12 weeks. Then, rats were all executed. Serum and liver tissues were obtained. Serum levels of TBIL, DBIL, ALT, AST, HA, LN, PCⅢ, TNF-α and the activities of SOD, GSH-Px and the contents of MDA, Hyp, MMP-1 and TIMP-1 in liver tissues were detected. Results: Results showed that total flavonoids of Hypericum japonicum could significantly alleviate liver fibrosis, as indicated by decreasing levels of TBIL, DBIL, ALT, AST, HA, LN, PCⅢ, TNF-α, Hyp, MMP-1 and TIMP-1. Moreover, total flavonoids of Hypericum japonicum could effectively enhance the activities of SOD, GSH-Px and decrease the content of MDA. Conclusions: Our results show that total flavonoids of Hypericum japonicum is effective in attenuating liver fibrosis induced by CCl4 composited factor in rats, at least partly, via enhancing the activation of antioxidant enzymes and inhibiting release of TNF-α.
