宝肾方对单侧输尿管结扎大鼠肾间质纤维化的保护作用 被引量:4

Protective effect of Baoshenfang on rats with renal interstitial fibrosis following unilateral ureteral ligation

摘要 目目的探讨宝肾方对肾间质纤维化的治疗价值及机制。方法将40只雄性Wistar大鼠行单侧输尿管结扎术,分为模型对照组和宝肾方小剂量组、中剂量组、大剂量组。按不同药物剂量,每天灌胃2次。另取10只大鼠为假手术组。第7、21、28天收集大鼠血清检测肝功能、肾功能,收集24 h尿液检测尿蛋白。第28天收集血清检测超氧化物歧化酶(SOD)和丙二醛(MDA),处死大鼠,取结扎侧肾脏,进行HE染色和Masson染色,观察肾小管间质纤维化程度。结果与假手术组相比,模型对照组大鼠血清肌酐(SCr)、血尿素氮(BUN)和24 h尿蛋白定量水平随着时间延长逐渐升高;第21、28天,不同剂量宝肾方组SCr、BUN和24 h尿蛋白定量均较模型对照组降低(P<0.05),宝肾方大剂量组SCr水平及尿蛋白定量低于宝肾方小剂量组(P<0.05)。与模型对照组比较,宝肾方组血清MDA含量降低,SOD活力升高。HE染色和Masson染色显示模型对照组及不同剂量宝肾方组肾间质纤维化相对面积较假手术组明显增加(P<0.05),不同剂量宝肾方组纤维化相对面积低于模型对照组(P<0.05)。结论宝肾方可明显减轻肾间质炎症细胞浸润及间质纤维化,延缓肾功能损害,宝肾方对单侧输尿管结扎大鼠肾小管间质纤维化有保护作用。 Objective To investigate the protective effect and mechanism of Baoshenfang on the renal interstitial fibrosis after unilateral ureteral obstruction(UUO) in rats. Methods 40 male Wistar rats treated with unilateral ureteral ligation were divided into the model group, the Baoshenfang low- dose group, middle- dose group and high- dose group. Rats received intragastric administration by Baoshenfang at different doses, 2times a day. The other 10 rats were sham group. Serum of rats were collected at 7th, 21 st, 28 th day after operation to test liver and kidney function, while 24 hours urine were collected to test urine protein. Superoxide dismutase(SOD)activity and malondialdehyde(MDA) level were tested from the serum collected at 28 th day.After the rats were sacrificed and the kidney tissues were collected for HE and Masson staining in order to observe the degree of renal interstitial fibrosis. Results Compared with the sham group, the levels of serum creatinine(SCr), blood ureanitrogen(BUN) and 24 h urine protein in the model group increased gradually. At 21stand 28 th day, the SCr, BUN and 24 h urine protein in different dose groups were lower than those in the model group(P〈0.05), and the levels of SCr and 24 h urine protein in the high-dose group were lower than those in the low- dose group(P〈0.05). Compared with the model group, the serum MDA levels in Baoshenfang groups were decreased, while the SOD activity was increased. HE staining and Masson staining showed that the relative area of renal interstitial fibrosis in the model group and the Baoshenfang groups was significantly increased compared with the sham group(P〈0.05), the relative area of the renal fibrosis in Baoshenfang groups was lower than that in the model group(P〈0.05). Conclusion Baoshenfang can significantly reduce the renal interstitial inflammatory cell infiltration and interstitial fibrosis, delay the damage to renal in UUO rats. Baoshenfang has protective effect on renal tubular interstitial fibrosis in rats with unilateral ureteral ligation.
