摘要
目的通过建立博莱霉素所诱导的大鼠肺间质纤维化模型,对肺纤维化模型大鼠病理形态、超氧化物歧化酶(SOD)、丙二醛(MDA)活性测定来探讨葛根素对实验性大鼠肺纤维化影响的作用机制。方法 SD雄性大鼠40只,随机分为对照组,模型组,葛根素组,激素组,每组10只,28d后,评价肺组织病理形态学变化,检测肺系数变化及各组肺组织匀浆中SOD活性和MDA含量。结果 (1)各组肺系数比较:第7、28d,模型组肺系数较其他3组明显增加(P<0.01),葛根素组与激素组肺系数比较无统计学意义(P>0.05)。(2)通过HE染色,观察到:葛根素与激素组的大鼠肺泡炎及肺纤维化程度均较模型组明显减轻,治疗7d后肺组织炎细胞、浸润区域较模型组明显减少;治疗28d后呈轻、中度肺纤维化改变,病变范围局限,肺泡间隔胶原纤维沉积呈束状。(3)葛根素与激素组较模型组大鼠肺组织匀浆SOD含量更高(P<0.01),MDA含量却降低(P<0.01),对照组大鼠肺组织匀浆SOD活性、MDA含量在各时间断基本一致。结论葛根素能减轻博莱霉素所致的大鼠肺泡炎及肺纤维化程度,提高肺组织SOD活力,降低MDA含量。
Objective To establish the Bleomycin-induced pulmonary fibrosis model in rats,explore the mechanism of the effects of puerarin on the pulmonary fibrosis in rats by the histopathology,the activity of the SOD,the contents of MDA. Methods Forty male SD rats were divided into four groups:control group(n=10),model group(n=10),puerarin group(n=10) and glucocorticoids group(n=10),On the 28 th day,the lungs were harvested for histophological examintion,the contents of superoxide dismutase(SOD) and malonyldialde-hyde(MDA) were detected. Results The comparison of the lung coeffcients:The lung coeffieients in the model group and in the other group were significantly(P〈0.01). Howevert there was no significance between puerarin group and glucocorticoids group(P〉0.05).(2)By the HE staining the alveolar inflammation and the Pulmonary fibrosis were more mitigated obviously in puerarin and glucocorticoids groups than model group;after treating for 7 days,pulmonary inflammation cells and infiltration area were more mitigated obviously in puerarin and glucocorticoids groups than in model Group;after treating for 28 days,the pulmonary fibrosis in puerarin and glucocorticoids group was changed lightly and moderately,the collagen fibers in pleural membrane and alveolar membrane were bunehy.(3)The contents of SOD in lung tissues homogenate of puerarin and glucocorticoids groups were higher than model group(P〈0.01);but the contents of MDA in lung tissues homogenate of puerarin and glucocorticoids groups were lower than model group(P〈0.01);during each period,the activities of SOD and the contents of MDA in lung tissues homogenate of control group were consistent basically. Conclusion Puerarin could significantly alleviate the degree of the alveolitis and pulmonary fibrosis,increase activity of SOD in lungs the rats with pulmonary fibrosis,and lower the content of MDA.
