摘要
为筛选适用于基因工程生产脂肪酶的微生物,从甘肃省兰州市区采集油脂污染土壤样品40份,通过分离培养,筛选出一株高产中性脂肪酶细菌。经形态学、生理生化和分子生物学鉴定,确定为枯草芽孢杆菌,命名为LZ-15。采用正交试验优化其产酶条件。结果表明,在蛋白胨1.5%、橄榄油2%、Mg SO4·7H2O 0.05%、(NH4)2SO40.1%、K2HPO40.2%、温度34℃、p H值7.0、接种量为7%的条件下,发酵液脂肪酶活力达7.58 U/m L,较优化前提高2.03倍。试验成功分离了一株脂肪酶高产枯草芽孢杆菌,并确定了其适宜的发酵生产条件。
For screening the microbe that could be used in genetic engineering to produce enzyme,forty samples of cultivated soil from Lanzhou city were collected,and a orthogonal experiment was designed to optimize the enzyme-producing conditions. The identification of colony morphology,microscopic morphology,physiological and biochemical and molecular biology indicated that the isolated microorganism was Bacillus subtilis,named LZ-15. The orthogonal experiment results showed that the lipase activity of strain LZ-15 reached 7.58 U/m L,under the condition of 1.5% peptone,2% olive oil,0.05% Mg SO4·7H2O,0.1%(NH4)2SO4,0.2% K2HPO4,fermentation temperature at 34℃,p H 7.0,7% inoculums volume,increasing the yield by 2.03 times.It has been concluded that we successfully isolated a bacillus subtitle with high lipase production,and the suitable incubation conditions are also certified.
