摘要
采用水提醇沉的方法提取芜菁粗多糖,经酸性乙醇分级和DEAE-52、Sephadex G-100纯化得芜菁多糖WJdp4-b。采用Sepharose CL-4B柱层析、HPLC法检测WJdp4-b的均一性,通过纸层析(PC)、气相色谱(GC)、红外光谱(IR)、核磁共振(NMR)、部分酸水解、高碘酸氧化和Smith降解、甲基化分析等分析方法对WJdp4-b的结构进行解析。结果表明:WJdp4-b为均一性多糖,相对分子质量约为10 840。其单糖组成为鼠李糖(Rha)、阿拉伯糖(Ara)、甘露糖(Man)、葡萄糖(Glc),各单糖的摩尔比为1.0∶1.0∶10∶9.0。结构分析表明:WJdp4-b为多分支结构,分子的主体是由Glc构成,连接方式为,Glc以1→3链接构成了WJdp4-b的主链,在Glc的6位处有分支,每17个Glc有12个分支。支链由Ara以1→4或1→5连接,Man的1→3、1→6连接构成,Rha和Glc构成了末端。
Crude polysaccharide was extracted from Brassica rapa L. After acidic ethanol fractionation,DEAE-52 and Sephadex G-100 gel filtration,WJdp4-bwas obtained. Its homogeneity is confirmed by chromatography on Sepharose CL-4B and HPLC. By means of HIO4 oxidation,Smith degradation,methylation,partial acid hydrolysis,paper chromatography,GC,IR,NMR,and GC-MS analysis,the structure of WJdp4-bwas identified. Its average molecular weight is 10 840. WJdp4-bwas composed of rhamose( Rha),arabinose( Ara),mannose( Man),glucose( Glc) and its molar ratio is 1∶ 1∶ 10∶ 9. Its main chain is mainly made up of( 1→4) Glc; Glc are substituted at 6-O and on average there were 12 branches among 17 main chain residues. The side chain was composed of( 1→4) or( 1→5)-Ara,( 1→3),( 1→6)-Man. The terminal residue was composed of Rha,Glc.
