摘要
目的研究人子宫内膜癌组织中是否存在血管生成拟态(VM),及其与血管内皮生长因子(VEGF)、环氧化酶-2(COX-2)表达的关系。方法收集子宫内膜癌标本60例及临床病理资料,CD34-PAS双重染色证实VM存在,分析VM与患者临床病理指标之间的关系。进一步对60例子宫内膜癌标本进行免疫组化技术检测VEGF、COX-2的表达水平并比较其在有VM组与无VM组之间的差异来探讨其在VM形成过程中的作用。结果 13例标本中存在VM,阳性率21.7%。具有VM的子宫内膜癌恶性程度高,易发生转移。VEGF、COX-2表达强度与子宫内膜癌的病理分级呈正相关(P<0.05)。存在VM的子宫内膜癌中VEGF、COX-2表达水平更高(P<0.05)。结论子宫内膜癌组织标本中存在VM;VM与子宫内膜癌的侵袭、转移密切相关;VEGF、COX-2在子宫内膜癌VM形成过程中发挥着重要调节作用。
Objective This experiment is to study whether vasculogenic mimicry is the special model of tumor blood vessels exists in en-dometrial carcinoma and the relationship of endometrial carcinoma vasculogenic mimicry with microvessel density VEGF,COX -2 expression. Methods Specimens of 87 cases were collected along with their clinicopathologic data. VM channels were detected using CD34/PAS double stai-ning and were then used to study the correlation between VM and clinicopathologic factors. Immunohistochemical staining were performed to these endometrial carcinoma patients,we studied the expression of VEGF,COX-2. Staining index(SI)was assessed to investigate the different ex-pression between VM-positive group and VM-negative group to study the effect in the formation of VM. Results 13 cases have VM of the all 60 patients(positive rate 21. 7%). The endometrial carcinoma cases belonging to VM group with higher invasive,had a higher rate of hematoge-nous metastases( P <0. 05). As the differentiated degree degrades,the expression of VEGF,COX-2 proteins was positive correlation( P <0. 05). The expression of VEGF,COX-2 proteins in the VM group was significantly higher than that in the non-VM group( P <0. 05). Conclu-sion VM exist in endometrial carcinoma. VM is related to invasion and metastasis of endometrial carcinoma. VEGF,COX-2 may play an impor-tant regulating role in the formation of VM in vasculogenic mimicry.
