摘要
为了阐明miR-103对牛骨骼肌卫星细胞的分化调节作用,试验将miR-103的过表达载体和miR-103抑制剂分别转染到分离培养的卫星细胞中,利用EDU功能检测试验、Real-time PCR和免疫荧光试验来分析miR-103对卫星细胞增殖和分化的调控作用,并用Western-blot和实时定量PCR检测miR-103对靶基因CCNE1的蛋白和mRNA水平表达情况的影响。结果表明:在牛骨骼肌卫星细胞中,miR-103能促进卫星细胞的分化并抑制细胞的增殖。说明miR-103是牛骨骼肌卫星细胞分化的一种重要的调控因子。
To clarify the role of miR - 103 in the differentiation and regulation on skeletal muscle satellite cells, the over - expression vector and the inhibitor of miR - 103 were transfected into the satellite cells which were isolated and cultured, respectively. EDU functional testing, Real - time PCR and immunofluorescence test were performed to analyze the role of miR - 103 in the regulation on the proliferation of the satellite cells, and then Western - blot analysis and Real - time quantitative PCR were used to detect the effect of miR - 103 on the expression levels of the protein and mRNA of the target gene CCNE1. The results showed that miR - 103 could promote the differentiation and inhibit the proliferation of satellite cells in the skeletal muscle satellite cells. The result indicates that miR - 103 is an important regulating factor in the differentiation of skeletal muscle satellite Ceils.
