摘要
目的建立同时测定西洋参保健食品中7种主要皂苷成分(人参皂苷Rg1、Rg2、Re、Rb1、Rb2、Rc和Rd)含量的高效液相色谱测定法(HPLc)。方法应用HPLC.梯度洗脱法,采用KromasilC18(4.6mm×250mm,5μm)色谱柱,以0.1%磷酸为流动相A,乙腈为流动相B进行梯度洗脱,流速1.0mL/min,检测波长203nm。结果人参皂苷Rg1、Re、Rb1、Rg2、Re、Rb2和Rd的检测浓度线性范围分别为0.056-0.56,0.203-2.03,0.324-3.24,0.105~1.05,0.048-0.48,0.104~1.04和0.152~1.52mg/mL,r值均大于0.999;平均加样回收率均大于98.7%,相对标准偏差(RSD)均小于1.9%(n=6)。结论此法能快速、准确地测定西洋参保健食品中7种主要皂苷的含量。
Objective To establish an HPLC method for determination of seven ginsenoside ingredients (ginsenoside Rg1, Rg2, Re, Rb1, Rb2, Rc and Rd) in health food of Panax quinquefolium. Methods An HPLC-gradient elution method was applied. The column was Kromasil C18 (4.6 mm ×250 mm, 5 μm), with 0.1% phosphoric acid solution as mobile phase A and acetonitrile as mobile phase B. The flow rate was 1.0 mL/min. The detection wavelength was set at 203 nm. Results The calibration curve was linear over the concentration range of 0.056-0.56, 0.203-2.03, 0.324- 3.24, 0.105-1.05, 0.048-0.48, 0.104-1.04 and 0.152-1.52 mg/mL for ginsenoside Rg1, Re, Rb1, Rg2, Re, Rb2 and Rd, respectively. The correlation coefficient of all curves were better than 0.999. The average recoveries of seven analytes were more than 98.7 %, and the RSD values were less than 1.9 % (n=6). Conclusion The developed HPLC method can be used for determination of seven ginsenoside ingredients in health food ofPanax quinquefolium.
