摘要
目的观察牛磺酸对四氯化碳(CCl4)诱导的小鼠肝纤维化模型血清中白介素(IL)-2的影响。方法将40只雄性SPF级C57B/L小鼠随机分为空白对照组、模型组、护肝片组和牛磺酸组,每组10只。空白对照组给予100%花生油1 ml/kg腹腔注射,模型组、护肝片组、牛磺酸组均给予含20%CCl4的花生油1 ml/kg腹腔注射,均注射10周。从第3周起护肝片组给予护肝片1 g/(kg·d),牛磺酸组给予牛磺酸500 mg/(kg·d)。第10周摘除眼球取血,检测小鼠血清中的透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、IL-2的含量;检测肝脏组织中超氧化物歧化酶(SOD)和丙二醛(MDA)的含量;计算肝脏指数;观察小鼠肝脏的病理组织学改变。结果模型组HA、LN、PCⅢ、Ⅳ-C、ALT、AST、IL-2、SOD和MDA的含量、肝脏指数与空白对照组相比具有显著性差异(P<0.05),在经过护肝片和牛磺酸治疗后,各组相关指标明显改善(P<0.05)。病理检查显示模型组肝细胞坏死并出现脂肪空泡、纤维组织增生和炎细胞浸润;护肝片组与牛磺酸组肝细胞结构完整,未见脂肪变与假小叶形成。结论牛磺酸可上调肝纤维化小鼠血清中IL-2的表达量,减轻CCl4诱导小鼠肝纤维化程度,改善肝功能。
Objective To observe the effect of Taurine on IL-2( interleukin-2) expression in Carbon Tetrachloride( CCl4) induced liver fibrosis in mice. Methods A total of 40 male SPF class C57B/L mice were randomly divided into control group,model group,Liver-aid tablets group and Taurine treatment group( n = 10 for each group). The control group was given intraperitoneal injection with 100% arachisoil( 1 ml / kg),while model,Liver-aid tablets and Taurine treatment groups were given arachisoil of 20% CCl4( 1 ml/kg),and all the groups were treated for 10 weeks. At the3 rdweek,Liver-aid tablets group was treated with 1 g /( kg·d) Liver-aid tablets,while Taurine treatment group was treated with 500 mg /( kg·d) Taurine. At the 10 thweek,blood samples were collected from mice by removing eyes,and the contents of serum hyaluronic acid( HA),laminin( LN),procollagen Ⅲ( PCⅢ),collagen-Ⅳ( Ⅳ-C),alanine transarninase( ALT),aspartate transarninase( AST) and interleukin-2( IL-2) were detected; the contents of superoxide dismutase( SOD) and malondialdehyde( MDA) in liver tissues were also detected; liver index was calculated; pathological and histological changes of livers in mice were observed. Results The differences in contents of HA,LN,PCⅢ,Ⅳ-C,ALT,AST,IL-2,SOD and liver index in model group were statistically significant compared with those in the control group( P 0. 05); related indicators improved significantly in all the groups after Liver-aid tablets and Taurine treatments( P 0. 05). Pathological examination showed that there were necrosis of liver cells,fat vacuoles,fibroplasia and inflammatory cell infiltration in model group; complete hepatocyte structures without fat change and pseudolobule in Liveraid tablets and Taurine treatment groups. Conclusion Taurine can increase the serum IL-2 expression and relieve liver fibrosis induced by CCl4 in mice with hepatic fibrosis.
