HPLC法同时测定杨梅叶中杨梅素等3种黄酮类化合物的含量 被引量:3

摘要 目的建立同时测定杨梅叶中杨梅素、槲皮素和山奈酚3种黄酮类化合物含量的反相高效液相色谱方法。方法采用Thermo Scientific ODS-2 Hypersil色谱柱(200mm×4.6mm,5μm);流动相为乙腈-体积分数0.2%甲酸水溶液,梯度洗脱;柱温为40℃;流速为0.7mL·min^-1;检测波长为360 nm。结果杨梅素、槲皮素和山奈酚的线性范围分别为15.0-150.0mg·L^-1、1.50-15.0mg·L^-1和1.0-10.0mg·L^-1,r≥0.999 4;平均加样回收率分别为100.2%、94.9%和104.2%,RSD分别为1.3%、1.0%和1.7%(n=6)。应用该法测定了3批杨梅叶中3种黄酮类化合物的含量。结论该方法准确、可靠、重复性好,为杨梅叶中黄酮类活性成分的开发利用提供参考。 Objective To establish an HPLC method for simultaneous determination of myricetin,quercetin and kaempferol in the leaves of Myrica rubra Sieb.et Zucc.Methods These paration was carried out on a Thermo Scientific ODS-2 Hypersil column(200mm × 4.6mm,5μm) with the mobile phase consisting of acetonitrile-0.2% formic acid under gradient elution.The column temperature was 40 ℃ and the flowrate was 0.7mL·min^-1,the detection wavelength was set at 360 nm.Results The linear ranges of myricetin,quercetin and kaempferol were 15.0-150.0mg·L^-1,1.50-15.0mg·L^-1and 1.0-10.0mg·L^-1,respectively.The average recoveries of myricetin,quercetin,kaempferol were 100.2%,94.9% and 104.2%,and the relative standard deviations(RSDs) were 1.3%,1.0% and 1.7%(n = 6).Conclusions This method is accurate,reliable and reproducible for quality control of leaves of Myrica rubra.
