摘要
目的通过观察SOX2和OCT4蛋白在乳腺纤维腺瘤和乳腺癌组织中的表达、分布,分析2种蛋白与乳腺癌的临床病理特征之间的关系,并探讨其在乳腺癌发生、发展和预后中的价值。方法应用免疫组织化学染色二步法,对40例乳腺纤维腺瘤和78例乳腺癌患者组织中的SOX2、OCT4的表达情况进行检测,应用统计学分析2种蛋白的表达和患者的肿块大小、年龄、临床分期以及组织学分级及淋巴结转移的关系。结果 140例乳腺纤维腺瘤组织中,有6例SOX2表达阳性,有4例OCT4表达阳性。278例乳腺癌组织中,SOX2和OCT4的阳性表达率是64%和87%,SOX2表达和组织学分级、淋巴结转移有关(P<0.05),而OCT4的表达和淋巴结转移、组织学分级以及肿瘤大小有关(P<0.05)。3SOX2、OCT4在乳腺癌中的表达明显高于乳腺纤维腺瘤组织,差异有统计学意义(P<0.05)。4乳腺癌的SOX2的异常表达强度随着OCT4表达强度的增强而升高,呈正相关(r=0.543)。结论 1SOX2和OCT4蛋白在乳腺癌中阳性表达率升高,反映出其表达情况在乳腺癌发生发展中起重要作用。2SOX2、OCT4的表达和乳腺癌在临床上的一些病理特征关系紧密,这2种蛋白的表达情况可以用来监测乳腺癌恶性程度。3SOX2与OCT4阳性表达,可能在乳腺癌的发生发展中起协同作用,这2种蛋白可以对乳腺癌的生物学行为进行准确预测,在制定个体化治疗方案中,也具有一定的临床价值。
Objective To determine the distribution and expression of SOX2 and OCT4 in breast fibroadenoma and breast carcinoma tissues, and to analyze the correlation between the two proteins and clinicopathological features of breast carcinoma, and to investigate the values of the two proteins in the occurrence, development and prognosis of breast carcinoma. Methods The expressions of SOX2 and OCT4 in tissue samples from 40 cases of breast fibroade-noma and 78 of breast carcinoma were determined by two-step immunohistochemical staining method. The correlation between the expressions of the two proteins and the tumor size, patient age, clinical staging, histological grade and lymph node metastasis was statistically analyzed, respectively. Results ①In tissue samples from 40 cases of breast fibroadenoma, 6 showed positive SOX2 expression, and 4 showed positive OCT expression. ②In 78 cases of breast carcinoma, the positive expression rate of OCT4 and SOX2 was 87% and 64%, respectively. The expression of SOX2 was significantly associated with histological grade and lymph node metastasis ( P〈0.05); whereas the expression of OCT4 was significantly associated with lymph node metastasis, histological grade and the tumor size ( P〈0.05). ③In breast carcinoma tissues, the expression levels of SOX2 and OCT4 were much higher than those in breast fibroadeno-ma tissues, with statistically significant differences (P〈0.05). ④ The intensity of abnormal expression of SOX2 in-creased along with higher expression intensity of OCT4 in breast carcinoma, with significantly positive correlation ( r=0.543). Conclusions ①The higher rates of positive expression of OCT4 and SOX2 suggest the important roles of the both in the occurrence and development of breast carcinoma. ②The expressions of OCT4 and SOX2 are significantly associated with certain clinicopathological features of breast carcinoma, and therefore can be used to monitor the de-gree of malignancy of breast carcinoma. ③The positive expressions of OCT4 and SOX2 may have a synergistic effect in the occurrence and development of breast carcinoma. The two proteins can be useful for accurate prediction of the biological behavior of breast carcinoma, and may be clinically valuable in individualized treatment.
