摘要
目的:验证岗梅提取物(LCE)对糖尿病大鼠溃疡的促愈合作用及对溃疡表面胶原、CD31(血小板内皮细胞黏附分子)表达的影响。方法:选取65只SD大鼠腹腔注射链脲佐菌素建立糖尿病模型,再结合皮肤缺损手术建立溃疡模型,随机将大鼠分为4个组,空白对照组、5%LCE软膏组、10%LCE软膏组以及Mupirocin阳性对照组。给药后检测溃疡愈合率,分别运用Masson三色染色法及免疫组化法检测CD31表达观察上皮组织胶原生长以及在溃疡愈合过程中的血管生成数目。结果:与空白对照组比较,LCE软膏组胶原合成量增多,创面愈合速度更快,且高剂量组创面愈合较低剂量组愈合更佳。免疫组化染色的结果显示10%LCE软膏组CD31的阳性表达量最多。结论:岗梅提取物可促进糖尿病大鼠溃疡的愈合,其机制与促进溃疡面的血管生成以及胶原合成有关。
Objective: To investigate the effects of ties asprella Champ extract (LCE) ointment on the wound healing process in diabetic rats and effects on collagen and CD3j expressions. Methods: 65 male SD rats were conducted and divided into four groups: blank group, 5% LCE group, 10% LCE group and mupirocin group. The diabetic model was induced by intraperitoneal injecting streptozotocin (STZ), then skin wound model was established by creating two cutaneous circular wounds for each rat at dorsal region. After different treat- ments, wound healing was assessed by rate of wound closure estimation. For histological study, Masson' s tri- chrome- stained was adopted to test the collagen formation. For further immunohistochemical study, CDal was employed to evaluate the angiogenesis. Results: LCE ointmentin could enhance the rate of wound closure and better collagen synthesis compared to blank control group, CD31 staining also showed more positive expressions which on behalf of the capillaries proliferation, and high - dose group showed better wound healing state than that of lower dose - group. Conclusion: LCE could significantly promote diabetic ulcer healing, and the preliminary mechanism might be associated with proliferation of capillaries and collagen synthesis.
