七氟烷吸入麻醉对大鼠认知功能影响的钙内流机制研究 被引量:2

摘要 目的:探讨七氟烷吸入麻醉对大鼠认知功能影响及细胞钙内流增强的参与作用。方法:SD大鼠随机分为4组:对照组、1.5%七氟烷吸入2h组(低剂量组)、3.0%七氟烷吸入2h组(高剂量组)及3.0%七氟烷吸入2h+钙通道阻滞剂硝苯地平组(硝苯地平组)。Morris型水迷宫测试各组大鼠认知功能,Western Blotting分析各组大鼠左侧海马IL-6/IL-8、TNF-α、细胞凋亡蛋白Bax/Bcl2及Caspase9的表达。结果:七氟烷组大鼠认知功能明显受到影响,IL-6/IL-8、TNF-α、细胞凋亡蛋白Bax及Caspase9表达明显上调,而Bcl2表达明显降低。硝苯地平组异常的认知功能及细胞凋亡蛋白得到明显恢复。结论:IL-6/8及细胞凋亡蛋异常表达及钙内流增强参与了七氟烷引起的大鼠认知功能障碍,硝苯地平可以明显恢复异常的大鼠认知功能障碍。 Objective: The present research aimed to explore the involvement of calcium influx in sevoflurane induced cognitive dysfunction during inhalation anesthesia. Methods.. SD rats were divided into 4 groups= control, 1.5% sevoflurane group, 3.0 % sevoflurane group and nifedipine group. The cognitive function and the expression of IL-6/IL-8, TNFα, Bax/Bcl2 and Caspase9 were detected by Western Blotting. Results= Cognitive function and Bcl2 expression were decreased and the ex- pression of IL-6/IL-8. TNFα, Bax and Caspase 9 was increased in 3.0 Msevoflurane group. Nifedipine normalized these abnor- malities greatly. Conclusion= IL6/IL8 and apoptotic protein as well as calcium influx were involved in sevoflurane induced cog- nitive function and nifedipinealleviates cognitive function effectively.
