TGF-β_1与自发性高血压大鼠肾脏功能关系的研究 被引量:9

RELATIONSHIP BETWEEN TGF-β_1 AND RENAL FUNCTION IN SPONTANEOUSLY HYPERTENSIVE RATS

摘要 探讨自发性高血压 (SHR)大鼠肾脏功能与转化生长因子 - β1的关系。方法 :研究组为SHR大鼠10只 ;同龄的WKY大鼠 7只为对照组。用ELISA法分别测定大鼠血清中TGF - β1的浓度 ;运用逆转录一聚合酶链反应 (RT -PCR)观察TGF -DmRNA在二组肾脏的表达 ;同时分别检测肾脏功能 (包括NAG酶 ,血肌酐 ,肌酐清除率 )。结果 :两组血清TGF - β1浓度的差异无显著性 (P >0 .0 5 ) ;SHR大鼠肾组织TGF - β1mR NA、尿NAG酶、血肌酐明显高于对照组 (P <0 .0 5 ) ,肌酐清除率明显低于对照组 (P <0 .0 5 ) ,肌酐清除率与肾脏局部的TGF - β1,mRNA的表达呈负相关 (P <0 .0 0 1) ;肾脏功能的改变与血清TGF - β1浓度高低无关。结论 :血压高低与血清TGF - β1浓度无关 ;肾脏局部TGF - β1mRNA的表达增高可能是引起高血压肾脏功能损害的重要因素。 Objective:To investigate the relationship between serum concentration of TGF-β 1 and blood pressure, and further to study the relationship between the TGF-β 1 and renal function. Methods:The serum concentration of TGF-β 1 was measured by ELISA, renal mRNA levels of TGF-β 1 were measured by RT-PCR, and the renal function including serum creatinine, urine NAG and Creatinine clearance were also measured.Results:The circulating levels of TGF-β 1 had no relationship with blood pressure ( P >0.05). Serum creatinine, urine NAG were positively correlated with TGF-β 1 mRNA ( P <0.01), Creatinine clearance was negatively correlated with TGF-β 1, mRNA ( P <0.01 ), and the renal function had nothing to do with the serum level of TGF-β 1( P >0.05) .Conclusions:The increased level of TGF-β 1 mRNA in renal tissue maybe an important factor of the damage of renal function in SHR.
