摘要
目的:观察高频、中频、低频电针心俞穴对垂体后叶素(PPI)诱导急性心肌缺血家兔模型的影响,探讨针刺心俞穴对家兔急性心肌缺血保护作用机理,为针灸临床治疗心血管疾病提供实验依据。方法:选择健康成年家兔随机分为对照组、模型组、高频、中频、低频电针组,电针选择针刺足太阳膀胱经心俞穴。通过PPI诱导急性心肌缺血模型,以MS-4000U生物信号采集系统记录Ⅱ导联心电图,分析第10 s、1 min、5 min、10 min、30 min、60 min时间点心率(HR)及ST-T变化。并采用生物化学方法测定血清肌酸激酶同功酶(CK-MB)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量。结果:心电图:模型组心率明显减慢,ST段改变显著,T波高耸,与对照组比较存在显著性差异;高、中频电针组心率有明显改善,高、中频电针组ST段及T波幅度改变有明显改善,与模型组比较存在显著性差异,低频组与模型组比较无显著性差异。血清CK-MB、SOD、MDA的变化:模型组血清CK-MB升高、SOD降低、MDA含量增加,与对照组比较存在显著性差异;高频、中频电针组血清CK-MB降低、SOD活性升高、MDA减少,与模型组比较差异存在显著性,低频电针组与模型组比较无显著性差异。结论:PPI诱导能够成功复制急性心肌缺血动物模型,各项观察指标变化明显。电针心俞穴对PPI诱导急性心肌缺血模型家兔的心电图ST-T有明显的改善作用。高、中频电针对PPI诱导家兔急性心肌缺血有明显保护作用,该作用与增强SOD的抗氧化能力、降低MDA含量以及降低心肌酶学水平有关。
Objective : To observe the protective effect of high - , mid - and low - frequency electro - acupunc- tures at Xinshu point on acute myocardial ischemia rabbits induced by PP1 and to provide experimental evidence for the treatment of cardiovascular diseases with acupuncture. Methods:We divided healthy adult rabbits into a control group, a model group, the high - , mid -and low- frequency electro -acupuncture groups. Electro -ac- upuncture was chosen at Xinshu point of bladder meridian. AMI model was established by PPI. MS -4000U bi- ological signal collection system recorded Ⅱ - lead electrocardiogram, whose T wave tower and ST shift of elec- trocardiogram were taken as acute myocardial ischemia index. That is to say, the model was successful repro- duction. Analyze HR and ST- T changes in each group of 10 s,1 min,5 min, 10 min,30 min and 60 min. Use biochemical methods to determine serum CK - MB, SOD and MDA contents. Results: Electrocardiogram: The slower HR and ECG showed towering T wave while ST shift significant in the model group. There was a statisti- cally significant difference compared with the control group. Compared with the model group ,the T wave ampli- tude,ST segment and HR of high - and mid - frequency groups were improved obviously. There was no statisti- cally significant difference between low - frequency electro - acupuncture group and model group. Serum : The contents of CK - MB,SOD and MDA in serum in the model group increased obviously, while SOD decreased significantly. There was a statistically significant difference compared with the control group. Compared with the model group, the content of SOD in serum increased obviously, while MDA and CK -MB decreased significantly in high -, mid - frequency electro - acupuncture groups. There was a statistically significant difference com- pared with the model group. There was no statistically significant difference between low -frequency electro - acupuncture group and model group. Conclusion:The animal model with acute myocardial ischemia induced by PPI can be successfully reproduced, and all the observed indexes change significantly. Electro - acupuncture at Xinshu point has an obviously improved effects on electrocardiogram ST - T of model rabbits with acute myocar- dial ischemia induced by PPI. High - and mid - frequency electro acupuncture has an obviously protective effect on AMI rabbits induced by PPI, which is associated with the strength of SOD antioxidant capacity, and the reduction of MDA content and myocardial enzyme level.
