参姜锁阳益气片对寒凝气滞血瘀证模型大鼠脑组织病理形态学和血脑屏障通透性的影响 被引量:1

Effect of Shenjiang Suoyang Yiqi Tablets on the Pathomorphology and Blood-brain Barrier Permeability in the Brain Tissues of Rats with Cold Coagulation Stagnation of Qi Blood Stasis

摘要 目的观察参姜锁阳益气片对寒凝气滞血瘀证大鼠脑组织形态学和血脑屏障通透性的影响。方法清洁级SD大鼠36只,随机选取6只为A组,其余30只采用肾上腺素加冰水浴复合低温冷冻法建立寒凝气滞血瘀证模型,造模成功后随机分为B、C、D、E、F组,每组6只。各组于末次造模后1 h灌胃给药,C、D、E组给予参姜锁阳益气片剂量分别为70、140、280 mg/kg,F组给予大株红景天胶囊280 mg/kg,A、B组灌胃给予等体积生理盐水,每日1次,连续2周。给药结束后,采用光镜和电镜观察脑组织病理变化,伊文思蓝灌注法检测大鼠脑组织血脑屏障通透性变化,干湿法测定脑组织水含量。结果镜下观察B组海马和下丘脑中神经细胞肿胀、排列紊乱,神经胶质细胞明显增多,毛细血管内皮细胞水肿严重,C、D、E、F组脑组织病理形态均有不同程度的改善。脑组织含水量和血脑屏障通透性B组高于A组,C、D、E、F组低于B组,C、D组高于F组(P<0.01)。结论参姜锁阳益气片能够有效抑制中枢神经系统功能异常,同时能够改善寒凝气滞血瘀大鼠海马及下丘脑组织病理学改变,从而对中枢神经系统起到一定的防护作用。 Objective To observe the effect of Shenjiang Suoyang Yiqi Tablets on the histomorphology and blood brain barrier permeability in the brain tissues of rats with cold coagulation stagnation of qi blood stasis. Methods A total of 36 SD rats were randomly divided into group A ( n = 6), cold coagulation stagnation of qi blood stasis models in the other 30 rats were established using adrenaline and ieewater bath method, and then the 30 rats were randomly divided into group B, C, D, E and F ( n = 6 for each group). On the 1 ~' h after model establishment, group C, D and E were respec- tively given Shenjiang Suoyang Yiqi Tablets of 70, 140 and 280 mg/kg; group F was given large Rhodiola capsules of 280 mg/kg; the group A and B received an equal volume of physiological saline. Continuous administration time was 14 d with ltime/d. After the last administration, the pathological changes of brain tissues were observed under light and elec- tron microscope, and permeability changes of blood brain barrier were detected using Evans blue perfusion method, and then wet and dry method was used to measure the water content of brain tissues. Results Pathological observation under microscope showed that nerve cells of hippocampus and hypothalamus tissues in group B were swollen, alinement in disor- der, obviously increased glial cells and badly edematous capillary endothelial cell, and pathological morphous of brain tis- sues in group D, E and F had improvement in different degrees. Compared with those in group B, water content of brain and permeability level of blood brain barrier in group A were lower, and the values in group C, D, E and F were also low- er, while the values in group C and D were higher than those in group F (P 〈 O. 01 ). Conclusion Shenjiang Suoyang Yiqi Tablets has certain protective effect on the central nervous system by effectively inhibiting the dysfunction of central nervous system, and improving the pathological changes in hippocampus and hypothalamus tissues of rats with cold coagu- lation stagnation of qi blood stasis.
