摘要
分别以D-氨基丁酸和DL-氨基丁酸为起始原料,经4步反应合成了右乙拉西坦和左乙拉西坦杂质A——(S)-N-(1-氨基-1-氧代丁基)-4-氯丁酰胺(纯度97.0%);左乙拉西坦经水解反应合成了左乙拉西坦酸(纯度99.9%),其结构经1H NMR和MS确证。
(R)-Levetiracetam and(S)-N-(1-amino-1-oxobutan-2-yl)-5-chloropentanamide with the purity of 97. 0% were synthesized by a four-step reaction from D-2-aminobutyric acid or 2-aminobutyric acid,respectively.(R)-α-ethyl-2-oxo-1-pyrrolidineacetic acid with the purity of 99. 9% was synthesized by hydrolysis of(S)-Levetiracetam. The structures were confirmed by^1 H NMR and MS.
