摘要
建立单核细胞增生李斯特氏菌实时浊度LAMP检测方法。根据单核细胞增生李斯特氏菌hly A基因设计4条特异性引物,采用环介导等温扩增方法 (LAMP)建立单核细胞增生李斯特氏菌快速检测方法。建立的LAMP方法具有高度特异性,经对14株细菌进行扩增,单核细胞增生李斯特氏菌为LAMP阳性,其他菌株均为LAMP阴性。建立的LAMP方法对培养的单核细胞增生李斯特氏菌的检测下限为32 CFU/m L,可在60 min内完成扩增反应。用建立的LAMP方法对160份不同食品进行检测,共检出5份LAMP阳性样本,与传统培养方法结果一致。该方法与传统方法相比,可节省大量时间,且对试验仪器和操作人员的要求低,具有良好的实用性。
In order to establish a loop-mediated isothermal amplification (LAMP) method for rapid diagnosis of Listeria monocytogenes, 4 primers which recognized the hlyA gene of Listeria monocytogenes was designed and used for LAMP assay. The results show that this method was high specific for Listeria monocytogenes. Through the amplification of 14 strains, it demonstrated that the Listeria monocytogenes was LAMP positive, and the other strains were LAMP negative. Furthermore this method cost only 60 minutes and the detection limit was 32 CFU/mL. Testing 160 seafood samples with the established method, the results indicated that the LAMP positive samples were 5, which accordance with the traditional methods. Compared to the traditional method, this method can save many times, low requirement of instruments and analyst, and has the good practicability.
