摘要
目的检测低氧诱导因子-1α(HIF-1α)和环氧合酶-2(COX-2)在寻常型银屑病皮损及正常人皮肤组织中的表达水平,探讨HIF-1α和COX-2在银屑病发病机制中的作用。方法收集30例寻常型银屑病和20例正常人的临床资料以及皮损和皮肤组织。(1)用半定量RT-PCR检测银屑病皮损和正常皮肤组织中HIF-1α和COX-2 m RNA表达情况;(2)用Western blot检测银屑病皮损和正常皮肤组织中HIF-1α和COX-2蛋白表达情况。结果 HIF-1α和COX-2 m RNA相对表达量在寻常型银屑病组(1.32±0.04和1.21±0.04)的表达显著高于正常对照组(0.49±0.03和0.31±0.03),两组相比差异有统计学意义(P<0.05);且HIF-1αm RNA相对表达量与COX-2 m RNA相对表达量呈正相关(r=0.664,P<0.05);HIF-1α和COX-2蛋白在寻常型银屑病组(0.76±0.03和0.62±0.03)的表达显著高于正常对照组(0.44±0.05和0.28±0.06),两组相比差异均有统计学意义(P<0.05);且HIF-1α蛋白表达量与COX-2蛋白表达量呈正相关(r=0.601,P<0.05)。结论 HIF-1α和COX-2在银屑病皮损中异常高表达且呈正相关,可能在银屑病的发生发展过程中起着重要作用。
ObjectiveTo study the expression and correlation of HIF-1αand COX-2 in samples of psoriasis vulgaris tissue and those of skin tissue which were from healthy individuals. To explore the role of HIF-1α and COX-2 in the pathogenesis of psoriasis.Methods 30 patients with psoriasis vulgaris and 20 normal human controls were enrolled into the study and obtained these lesions or normal skin tissues. (1)RT-PCR was used to examine the mRNA expressions of HIF-1α and COX-2 in samples of psoriasis vulgaris lesion tissues and those of skin tissues from healthy individuals. (2)Western blot was used to examine the protein expressions of HIF-1α and COX-2 in samples of psoriasis vulgaris lesion tissues and those of skin tissues from healthy individuals.Results The expressions of HIF-1α and COX-2 mRNA in psoriasis vulgaris tissues (1.32±0.04 and 1.21±0.04) were significantly higher than those in the normal group (0.49±0.03 and 0.31±0.03). Which showed significant difference in the expressions of HIF-1α and COX-2 mRNA between the psoriasis and the control group (P〈0.05). Moreover, The HIF-1αmRNA in psoriatic lesions was associated significantly with COX-2 mRNA (r=0.664,P〈0.05). The expressions of HIF-1α and COX-2 protein in psoriasis vulgaris tissues (0.76±0.03 and 0.62±0.03) was significantly higher than those in the normal group (0.44±0.05 and 0.28±0.06). Which showed significant different in the expressions of HIF-1α and COX-2 protein between the psoriasis and the control group(P〈0.05); and HIF-1α protein in psoriatic lesions was associated significantly with COX-2 protein (r=0.601,P〈0.05).ConclusionHIF-1α and COX-2 high expression and their positive correlation in psoriatic lesions might play an important role in genses and development of psoriasis.
