摘要
目的研究髓源性抑制细胞(myeloid derived suppressor cells,MDSC)与鼻咽癌细胞上皮-间质转化(epithelialmesenchymal transition,EMT)的关系及对鼻咽癌细胞侵袭转移的影响。方法用流式细胞技术检测16例鼻咽癌患者以及9例健康对照者骨髓中MDSC所占比例,并分离鼻咽癌组织中MDSC;将MDSC与鼻咽癌细胞株CNE-2共培养,观察CNE-2细胞形态学的改变;应用q RT-PCR技术从RNA水平检测共培养前后CNE-2细胞上皮-间质转化相关基因(E-cadherin,N-cadherin,Vimentin,Snail)的改变;进一步采用Western blot技术从蛋白水平检测其表达变化;继而通过transwell小室及划痕实验检测MDSC对CNE-2细胞侵袭转移能力的影响。结果鼻咽癌患者骨髓中MDSC所占比例(10.37%)明显高于健康对照者(1.65%);MDSC与CNE-2细胞共培养后,CNE-2细胞在形态学上发生EMT;q RT-PCR和Western blot分别从RNA和蛋白水平证实CNE-2细胞上皮表型的相关基因(E-cadherin)表达下调,间质表型的相关基因(N-cadherin,Vimentin,Snail)表达上调;transwell小室侵袭实验证实共培养后CNE-2细胞的侵袭能力增强;划痕实验证实共培养后CNE-2细胞的迁移能力增强。结论 MDSC通过诱导EMT促进鼻咽癌细胞的侵袭转移能力。
To study the relationship between myeloid-derived suppressor cells(myeloid derived suppressor cells, MDSC) and nasopharyngeal carcinoma cells epithelial cells-mesenchymal transition(EMT) and its impact on the invasion and metastasis of nasopharyngeal carcinoma cell, MDSC proportion of bone marrow from 16 cases of nasopharyngeal carcinoma(NPC) patients and nine healthy controls were detected by flow cytometry. Also the MDSC was co-cultured with nasopharyngeal carcinoma cell line CNE-2, and morphological changes of CNE-2 was observed; q RT-PCR technology was used to detect the RNA changes of EMT related genes(E-cadherin, N-cadherin, Vimentin, Snail) in the training CNE-2 cells before and after the co-cultivation, further, Western blot technique was applied to detect the the changes in protein level. Then transwell chamber and scratch assay were used to evaluate the effects of MDSC on the invasive ability of CNE-2 cells. Data showed that the MDSC percentage in bone marrow of patients with nasopharyngeal carcinoma was 10.37%, which was significantly higher than that in healthy controls(1.65%); After co-cultivation with MDSC, CNE-2 cell EMT occurred morphologically; q RT-PCR and Western blot indicated E-cadherin downregulated, while N-cadherin, Vimentin and Snail upregulated in RNA and protein levels. Furthermore, MDSC enhanced the invasion and migration of CNE-2 cell. In conclusion, MDSC can promote invasion and metastasis of nasopharyngeal carcinoma cells by inducing EMT.
