摘要
以凤丹白牡丹(Paeonia ostii cv.‘Fengdaibai’)果荚为原料,研究了牡丹果荚多糖的提取工艺以及牡丹果荚多糖的抗氧化活性。在单因素试验的基础上,根据Box—Behnken试验设计的原理,选取提取温度、提取时间和液固比例三因素三水平进行中心组合试验,通过响应面分析得到优化的牡丹果荚多糖提取组合条件。结果表明提取最佳工艺条件为原料粒度60~80目、提取温度100℃、提取时间40min、液固比例20mL·g-1,并采用终浓度为80%的乙醇进行沉淀析出。在此条件下,牡丹果荚多糖的得率预测值为8.76%,验证值为8.77%。采用DPPH和还原力体系检测了牡丹果荚多糖的体外抗氧化能力,结果显示牡丹果荚多糖具有一定水平的抗氧化能力。
We optimized the polysaccharide extraction from the peony pod and studied its antioxidant activity. By single factor design, we identified extraction parameters of particle size of material, extraction temperature, extraction time, liquid-solid ratio and final concentration of ethanol in precipitation step. A 3-factor, 3-level central composite design involving three independent variables followed by response surface analysis was performed for the further optimization of the extraction parameters. The optimal extraction conditions were 60 - 80 mesh of particle size, extraction temperature of 100℃, extraction time of 40 min, liquid-solid ratio of 20 mL · g-1, and precipitated ethanol with the final concentration of 80%. Under these conditions, the predicted and experimental extraction yields of pod polysaccharides were 8.76% and 8.77%, respectively. The antioxidant capacity of pod polysaeeharide was determined by DPPH and reducing activity. The pod polysaccharide has the antioxidant capacity.
