摘要
目的为了简化布鲁杆菌试管凝集试验(SAT)操作繁琐、取液不精确的问题,改进试管凝集试验的操作方法。方法用微量移液器取代吸管,被检血清200μl减少到40μl,血清稀释度从1管-5管分别为1∶25、1∶50、1∶100、1∶200和1∶400精简为4个稀释梯度,分别为1∶50、1∶100、1∶200和1∶400,精简操作步骤,用此改进方法和传统操作方法同时检测虎红筛查阳性血清样品97份。结果改进方法和传统操作方法检测结果符合率达到98.91%,而两种SAT与RBPT检出的阳性符合率分别为达到93.81%和94.85%,改进的试验方法具备操作简便,试验结果客观、精确,血清用量小等优点。结论建议用改进的试管凝集试验方法取代传统的试管凝集试验方法。
Objective To simplify complicated operation and liquid measuring uncertainty of brucella serum tube agglutination test (SAT), and to improve the operation methods of serum tube agglutination test. Methods Using micropipettor to replace straw, detected serum decreased from 200 μl to40 Ixl, serum dilution from 1 to5 were 1 : 25, 1 : 50, 1 : 100, 1 : 200 and 1 : 400 reduced to 4 dilution gradient, respectively 1 : 50, 1 : 100, 1 : 200 and 1 : 400, reduce and simplify the operations, and detect rose bengal at the same time for screening positive serum 97 samples by using the improved method and the traditional method. Results The rate of coincidence on operation test results with the improved method and the traditional method reached 98.91%, and SAT and RBPT positive rate were 93.81% and 94.85 %, improved test method has simple operation, objective, accurate test results, small amount of serum etc. Conclusion Recommend the improved method to replace the traditional oper- ation method.
