摘要
丙酮酸是细胞质糖代谢的最终产物,拥有多种代谢途径。多年来科学家们对线粒体展开了广泛的研究,发现存在一种特殊的线粒体丙酮酸载体(MPC),它在丙酮酸转运中发挥重要作用。肿瘤细胞偏向有氧糖酵解获取能量,即瓦伯格效应。理论上MPC功能的降低应该与肿瘤细胞的瓦伯格效应相关。目前,利用特异性抑制剂已经实现了MPC的纯化,并进一步鉴定认识到该蛋白的结构和功能。然而,由于MPC在人类细胞中的存在近期才被阐明,这方面的研究还很有限。
Pyruvate is the final product of glycometabolism in cytoplasm. Through decades of effort, scientists have recently characterized the special mitochondirial pyravate carrier(MPC) ,an important role in the mitochondrial transport of pyruvate. Tumor cells are known with Warburg effect, i. e. aerobic glycolysis. We speculate that suppression of MPC expression or MPC function may be linked to the Warburg effect. Nowadays, MPC can be purified by the specific inhibitors, thus the structure and function of this protein can be identified. However the studies of MPC in tumor cells are limited, mainly because of the fact of failure in characterization of MPC in human cells.
