摘要
目的探讨汉防己甲素对大鼠早期实验性矽肺的治疗效果。方法取健康成年大鼠90只按随机数字表分成汉防己甲素组、阳性对照组(同期染尘)、阴性对照组(不染尘),每组30只。用石英粉尘注入大鼠气管,制作矽肺动物模型。染尘后第4天开始给药,汉防己甲素组给予汉防己甲素灌胃,另两组给予等量超纯水灌胃。治疗结束后分别处死大鼠,进行全肺干湿重称重、羟脯氨酸含量测定、支气管肺泡灌洗液细胞计数及分类、病理组织学检查。结果各组大鼠体重随年龄的增长呈上升趋势(P<0.05),但组间体重变化趋势程度不同;染尘后,汉防己甲素组体重比阳性对照组重(P<0.05)。汉防己甲素组大鼠的全肺干湿重均低于阳性对照组(P<0.05)。染尘第50天时,汉防己甲素组大鼠羟脯氨酸含量与阴性对照组差异无统计学意义(P>0.05),而阳性对照组高于阴性对照组(P<0.05)。病理组织学提示汉防己甲素组肺实变程度低于阳性对照组。结论汉防己甲素对大鼠早期实验性矽肺有一定的改善作用。
Objective To evaluate the efficacy of sinomenine on rats with early stage experimental silicosis . Methods Ninety healthy adult rats were randomly divided into sinomenine group,positive control group(exposed to dust concurrently),and negative control group ( not exposed to dust ) ,with 30 rats in each group .The animal model of silicosis was prepared by priming quartz into trachea of rats .The treatment began on the fourth day after exposure to dust .The sinomenine group was given sinomenine by gavage .The positive control group and negative control group were given pure water by gavage .The rats were sacrificed when the therapies were over .The lungs of rats were weighed ,hydroxyproline content was measured ,bronchoalveolar lavage fluid cells were counted and classified ,and pathological examination was performed .Results The weights of the rats rose with increasing ages ( P〈0.05 ) , while the weigh variations showed different degrees among groups .The weight of sinomenine group was heavier than that of positive control group after exposure to dust(P〈0.05).The whole lung dry-wet weight of sinomenine group was lower than that of positive control group (P〈0.05).On the 50th day after exposure to dust ,the hydroxyproline content showed no significant difference between sinomenine group and negative control group (P〉0.05).However,the hydroxyproline content of positive control group was higher than that of negative control group (P〈0.05).The pathological examination showed that the degree of lung consolidation in the sinomenine group was less than that in the positive control group .Conclusion Sinomenine has a certain improvement on rats with early stage experimental silicosis .
