摘要
目的探讨姜黄素对大鼠血管平滑肌细胞单核细胞趋化蛋白-1表达、增殖和凋亡的影响。方法分离SD大鼠血管平滑肌细胞进行培养传代,采用不同浓度的姜黄素作用于氧化型低密度脂蛋白(Ox-LDL)诱导的大鼠平滑肌细胞,根据作用药物浓度进行分组,对照组(不加任何药物),Ox-LDL 100μg/mL组,Ox-LDL 100μg/mL+姜黄素20μmol/L组,Ox-LDL 100μg/mL+姜黄素40μmol/L组,Ox-LDL 100μg/mL+姜黄素80μmol/L组。分析姜黄素对大鼠血管平滑肌细胞单核细胞趋化蛋白-1(MCP-1)的表达、对细胞的增殖的影响。按照加入药物浓度进行分组,对照组(不加任何药物),姜黄素50μmol/L组,姜黄素100μmol/L组,姜黄素120μmol/L组,分析姜黄素对细胞凋亡的影响。结果 Ox-LDL 100μg/mL组MCP-1分泌[(812.6±78.7)ng/L]及MCP-1 mRNA表达水平[(1.18±0.11)]较对照组[(91.3±6.1)ng/L,(0.16±0.04)]显著升高,差异有高度统计学意义(P<0.01)。姜黄素对OxLDL诱导的大鼠血管平滑肌细胞MCP-1分泌及MCP-1 mRNA表达、细胞增殖具有显著的抑制作用,对细胞凋亡有诱导作用,并且随着姜黄素浓度的增加,作用越明显(P<0.01)。结论姜黄素能够抑制Ox-LDL诱导的大鼠细胞平滑肌细胞MCP-1表达以及细胞增殖,诱导大鼠细胞平滑肌细胞凋亡。
Objective To discuss effect of curcumin on MCP-1,cell multiplication and apoptosis in Ox-LDL inducing rat VSMCs. Methods VSMCs of rats were isolated culture. Different doses of curcumin actted on Ox-LDL inducing rat VSMCs. Rats were divided into control group（without any drug）, Ox-LDL 100 μg/mL group, Ox-LDL 100 μg/mL＋curcumin 20 μmol/L group, Ox-LDL 100 μg/mL＋curcumin 40 μmol/L group, Ox-LDL 100 μg/mL＋curcumin 80 μmol/L group according to the concentration of the drug. MCP-1 expression, and cell multiplication were detected. Rats were divided into control group（without any drug）, curcumin 50 μmol/L group, curcumin 100 μmol/L group, curcumin 120μmol/L group according to the concentration of the drug, and apoptosis were detected. Results MCP-1 excreting[（812.6±78.7） ng/L] and MCP-1 mRNA expression [（1.18±0.11）] of Ox-LDL 100 μg/mL group were higher than those of control group [（91.3±6.1） ng/L,（0.16±0.04）], the differences were statistically significant（P〈0.01）. Curcumin showed inhibition on MCP-1 excreting, MCP-1 mRNA expression, cell multiplication in Ox-LDL inducing rat VSMCs,and induction on apoptosis. Those action showed stronger with enrichment（P〈0.01）. Conclusion Curcumin shows inhibition on MCP-1 excreting and cell multiplication in Ox-LDL inducing rat VSMCs, and shows induction on apoptosis of rat VSMCs.
