补肾利湿法中药复方对大鼠痛风性关节炎NALP6信号传导通路调控作用的研究 被引量:9

Study on regulatory effect of Chinese herbal compound for invigorating kidney and removing dampness on NALP6 signaling pathway in rats with gouty arthritis

摘要 目的探讨补肾利湿法中药复方对痛风性关节炎大鼠NALP6信号传导通路的调控作用。方法将84只SD大鼠随机分成空白对照组、模型组、中药痛风舒组、西药秋水仙碱组、中药低剂量组、中药中剂量组、中药高剂量组。每天1次进行药物灌胃。在第7天给药1 h后,采用改良复合氧嗪酸钾和尿酸钠造模方法制作痛风性关节炎大鼠模型。造模4 h后取材,观察各组大鼠滑膜组织病理变化,采用ELISA法检测血清血尿酸、caspase-1、IL-1β、IL-18的含量,并以Western blot法检测滑膜组织内NALP6蛋白的表达水平。结果经补肾利湿中药复方干预的中药中剂量组,其大鼠滑膜组织病理学改变减轻,体内血尿酸值明显下降,血清caspase-1、IL-1β和IL-18表达水平显著下降,与模型组比较,差异有统计学意义(P<0.05);经补肾利湿中药复方干预的中药高剂量组,Western blot检测滑膜组织内NALP6蛋白的表达明显下调,与模型组比较,差异均有统计学意义(P<0.05)。结论阻断NALP6-caspase-1-IL-1β这一炎症信号通路是补肾利湿中药复方防治痛风性关节炎的重要机制之一。 Objective To investigate the regulatory effect of Chinese herbal compound for invigorating the kidney and removing dampness on the NALP6 signaling pathway in rats with gouty arthritis(GA).Methods Eighty-four SD rats were randomly assigned into the blank control group,model group,tongfengshu group,colchicine group,low-dose Chinese medicine group,medium-dose Chinese Medicine group and high-dose Chinese Medicine group.Gavage administration was performed once daily.At 1h after administration on day 7,the modified composite modeling method with potassium oxazinate and sodium urate was used to establish the GA rat model.Samples were collected at 4h after modeling,the pathological changes in synovial tissues were observed in rats of each group,the content of uric acid,caspase-1,IL-1βand IL-18 in serum was determined by ELISA and the expression level of NALP6 in synovial tissues was detected by Western blot.Results In the medium-dose Chinese medicine group treated with Chinese herbal compound for invigorating the kidney and removing dampness,the pathological changes in synovial tissues,the value of blood uric acid in the body,and the expression levels of caspase-1,IL-1βand IL-18 in serum decreased significantly compared to the model group(P<0.05);and in the high-dose Chinese medicine group treated with the Chinese herbal compound,Western blot analysis showed that the expression level of NALP6 decreased significantly in synovial tissues compared to the model group(P<0.05).Conclusion Blocking the inflammatory signaling pathway of"NALP6-caspase-1-IL-1β"is an important mechanism of action in preventing and treating gouty arthritis with Chinese herbal compound for invigorating the kidney and removing dampness.
