摘要
目的:探讨抗纤灵方对5/6肾切除诱导肾纤维化模型大鼠肾功能及细胞外基质(extracellular matrix,ECM)表达的影响。方法:将50只SD大鼠随机分为正常组(n=7),假手术组(n=7),手术组(n=36)。手术组建立5/6肾切除肾纤维化模型,将造模成功的大鼠随机分为模型组、抗纤灵组、氯沙坦钾组。氯沙坦钾组、抗纤灵方组每日分别给予氯沙坦钾(33.3g·kg^-1)及抗纤灵方(21g·kg^-1)灌胃治疗,其余正常组、假手术组、模型组都用等体积的生理盐水干预,24周后测定各组肾功能,24h尿量及24h尿蛋白定量,免疫组化测定肾脏组织Ⅰ型胶原(ColⅠ),Ⅲ型胶原(ColⅢ)表达情况,蛋白免疫印迹法(Westernblot)测定肾脏组织ColⅠ,ColⅢ蛋白表达水平。结果:与正常组和假手术组比较,模型组血肌酐,尿素氮,24h尿量,24h尿蛋白定量及肾组织ColⅠ,ColⅢ表达水平均明显升高(P<0.01);与模型组比较,抗纤灵方组和氯沙坦钾组血肌酐,尿素氮,24h尿蛋白及肾组织ColⅠ,ColⅢ蛋白表达水平显著下降(P<0.05,P<0.01)。结论:抗纤灵方能下调肾脏组织ColⅠ,ColⅢ表达,从而抑制ECM的积聚,发挥延缓肾脏纤维化和保护肾功能的作用。
Objective: To investigate the effect of Kangxianling decoction on renal function and expression of extracellular matrix(ECM) in renal tissue of rats with renal fibrosis induced by 5/6 nephrectomy.Method: Totally 50 SD rats were randomly divided into control group(n = 7),sham-operation group(n = 7),operation group(n = 36). The 5/6 nephrectomized renal fibrosis model was established in operation group. After successful modeling,rats were randomly divided into model group,Kangxianling group and losartan potassium group. The losartan potassium group and the Kangxianling group were given losartan potassium(33. 3 g·kg-1) and Kangxianling decoction(21 g·kg-1) every day. The control group,sham-operation group and model group were all treated with equal volume of normal saline. Rats were put to death after 24 weeks of consecutive medication,and renal function,24 h urine volume and 24 h-Pro quantitation of each group were measured. The expressions of collagen Ⅰ(Col Ⅰ) and Col Ⅲ in renal tissues were determined by immunohistochemistry and Western blot.Result: Compared with the normal group and sham-operation group,serum creatinine(SCr),blood urea nitrogen(BUN),24 h urine volume,24 h-Pro quantitation,renal tissue Col Ⅰ and Col Ⅲ expression levels were significantly increased in the model group(P < 0. 01). Compared with the model group,the levels of SCr,BUN,24 h-Pro quantitation,protein expression levels of renal tissue ColⅠ and Col Ⅲ were significantly decreased in the Kangxianling group and the losartan potassium group(P < 0. 05,P < 0. 01). Conclusion: Kangxianling decoction can down-regulate the expressions of Col Ⅰ and Col Ⅲ in kidney tissue,there by inhibiting the accumulation of ECM,and exerting the effect in delaying renal fibrosis and protecting renal function.
