摘要
目的鉴定福建省2016-2017年导致儿童腹泻的新型诺如病毒,并分析其分子特征。方法收集2016-2017年福建省病毒性腹泻监测哨点医院5岁以下腹泻住院儿童粪便标本,共计892份。应用RT-PCR方法检测诺如病毒,对诺如病毒RNA聚合酶、衣壳蛋白片段进行序列测定和分子流行特征分析。结果892份标本检出诺如病毒核酸阳性141份,其中GⅠ型6份,GII型125份,GⅠ和GII混合型10份。107份GII型诺如病毒标本成功测序。序列分析结果表明,主要型别为GII.4型和GII.3型,且检出多种新型诺如病毒,包括GII.P17/GII.17、GII.P16/GII.2和GII.P16/GII.4,毒株的核苷酸序列分别与2014年日本的Kawasaki323株、2016年日本的Kawasaki151毒株和2016年英国的NOR2565株高度同源。结论福建省腹泻儿童检出多种新型诺如病毒,需密切关注其流行趋势。
ObjectiveTo identify and to analyze the molecular characteristic of novel norovirus in children with diarrhea in Fujian province in 2016-2017.MethodsA total of 892 stool samples were collected from hospitalized children under 5 years old with diarrhea in 2016-2017. Norovirus was detected using the reverse transcription-PCR (RT-PCR). The RNA dependent RNA polymerase (RdRp) gene and capsid gene were partially sequenced, then their molecular epidemiology characteristics were analyzed.ResultsFrom 892 samples, nucleic acid of norovirus was positive in 141 samples, including 6 sampels with genogroup I (GI), 125 samples with genogroup II (GII) and 10 samples with mixed infections of GI and GII. GII norovirus in 107 samples were successfully sequenced. Sequence analysis indicated that GII. 4 and GII. 3 were the major genotypes. Various novel genotypes of norovirus were also identified, including GII. P17/GII. 17, GII. P16/GII. 2 and GII. P16/GII. 4. The strains had highly nucleotide identities with Kawasaki323 strain in Japan in 2014, Kawasaki151 strain in Japan in 2016 and NOR2565 strain in England in 2016, respectively.ConclusionsNovel genotypes of norovirus were detected among children with diarrhea in Fujian province. The epidemic trends of the viruses should be paid close attention.
