摘要
目的建立测定丙泊酚中/长链脂肪乳注射液与含葡萄糖的注射液配伍时5-羟甲基糠醛含量的高效液相色谱(HPLC)法。方法采用十八烷基硅烷键合硅胶为填充剂(ACE Excel Super C18,250 mm×4.6 mm,5μm),以甲醇-0.5%醋酸水(10∶90,V/V)为流动相;流速1.0 m L·min^(-1);柱温30℃;检测波长284 nm。结果 5-羟甲基糠醛在0.01~8.57μg·mL^(-1)浓度范围内与峰面积呈良好的线性关系(r=1. 000 0),精密度和重复性的RSD分别为1.2%和0.54%。结论方法操作简便,结果准确可靠,可作为丙泊酚中/长链脂肪乳注射液与含葡萄糖注射液配伍时5-羟甲基糠醛含量的测定方法。
OBJECTIVE To establish a method for the determination of 5-hydroxymethylfurfural (5-HMF)when propofol medium/long-chain fat emulsion injection is mixed with glucose injection.METHODS HPLC was carried out on ODS-C18 column (ACE Excel Super C18 ,250mm×4.6mm,5μm),with methanol-0.5% acetic acid (10:90,V/V)as the mobile phase.The flow rate was 1.0mL·min^-1,and the column temperature was set at 30℃ with the detection wavelength of 284nm.RESULTS The standard curve was in good linearity in the range of 0.01-8.57μg·mL^-1 for the determination of 5-HMF (r=1.0000).Precisions and repeat abilities of the methods were 1.2% and 0.54% respectively.CONCLUSION The method is simple,reliable and suitable for the quality control of 5-HMF when propofol medium/long-chain fat emulsion injection is mixed with glucose injection.
