摘要
本研究采用DNA条形码技术对黑龙江流域的彩石鳑鲏(Rhodeus lighti)、方氏鳑鲏(Rhodeus fangi)和黑龙江鳑鲏(Rhodeus sericeus)进行了系统进化和遗传多样性分析。单倍型检测发现,3种鳑鲏的73条序列中共检测到9种单倍型,其中黑龙江鳑鲏独享5种单倍型,彩石鳑鲏和方氏鳑鲏各有3种单倍型,二者共享2种单倍型。遗传多样性分析显示,彩石鳑鲏和方氏鳑鲏遗传多样性远低于黑龙江鳑鲏,这表明彩石鳑鲏和方氏鳑鲏的种群内遗传分化不大。通过与GenBank公开发表的10种鳑鲏属鱼类mtDNACOⅠ序列聚类分析发现,3种鳑鲏均与已发表同种序列聚在一起,黑龙江鳑鲏单独聚为一支;彩石鳑鲏与粗纹暗色鳑鲏、高体鳑鲏和中华鳑鲏聚为一支;方氏鳑鲏与济南鳑鲏和暗色鳑鲏聚为一支。针对以上结果,本研究对分布在中国黑龙江流域的黑龙江鳑鲏、彩石鳑鲏和方氏鳑鲏3个种群遗传多样性和中国鳑鲏亚科鱼类的进化关系进行了探讨和梳理,为其属内种间系统发育关系的进一步研究及其自然分类地位提供一定的理论依据和基础资料。
The phylogeny and genetic diversity of Rhodeus lighti, Rhodeus fangi and Rhodeus sericeus in Heilongjiang river were analyzed using DNA barcoding in this study. Through haplotype detection, a total of 9 haplotypes were found in 73 sequences of 3 kinds of Rhodeus, of which five were for Rhodeus sericeus, three for Rhodeus lighti and Rhodeus fangi, respectively, and the latter two shared two haplotypes. The genetic diversity analysis showed that Rhodeus lighti and Rhodeus fangi were much lower than that of Rhodeus sericeus, which indicated that there was low genetic differentiation in the populations of Rhodeus lighti and Rhodeus fangi. Through clustering analysis for mtDNA COⅠsequences from ten Rhodeus species deposited in GenBank, we found Rhodeus lighti, Rhodeus fangi and Rhodeus sericeus clustered with the same published sequences. The Rhodeus sericeus clustered alone, Rhodeus lighti clustered with Rhodeus suigensis, Rhodeus ocellatus clustered with Rhodeus sinensis, and Rhodeus fangi clustered with Rhodeus notatus and Rhodeus atremius. In view of the above results, the genetic diversity for Rhodeus lighti, Rhodeus fangi, Rhodeus sericeus distributed inHeilongjiang river and the evolutionary relationship for Chinese Rhodeinae species were discussed and sorted out, which could provide certain theoretical basis and basic data for the further study on its phylogenetic relationship in the genera level and species level and its natural classification status.
