摘要
目的:探讨电针联合骨髓间充质干细胞(BMSCs)移植对脊髓损伤(SCI)大鼠炎症反应及损伤组织神经营养因子表达水平的影响。方法:50只健康成年SD大鼠随机分为正常组、模型组、电针组、BMSCs组与联合组,各10只,除正常组、模型组外,其他各组于SCI后1周开始给予电针、BMSCs或联合治疗,共干预12周,治疗后第4、8、12周通过BBB评分评价各组大鼠肢体运动功能,经由Western-blot定量检测损伤脊髓组织白介素-6 (IL-6)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)蛋白表达水平;通过免疫组化法测定治疗后第4、12周脑源性神经营养因子(BDNF)、神经生长因子(NGF)表达水平。结果:与正常组比较,其他各组大鼠第4、8、12周BBB评分,第4、12周BDNF、NGF表达水平均显著低,差异有统计学意义(P<0. 05);电针组、BMSCs组、联合组大鼠第4、8、12周BBB评分,第4、12周BDNF、NGF表达均显著高于模型组(P <0. 05);联合组不同时间点BBB评分、BDNF、NGF表达水平均显著高于电针组、BMSCs组(P<0. 05);电针组、BMSCs组、联合组第4、8周IL-6、TNF-α、IL-1β蛋白表达均显著低于模型组(P<0. 05)。结论:电针联合BMSCs移植可能通过抑制SCI大鼠炎症反应,增强损伤脊髓神经营养因子、神经生长因子以促进大鼠运动功能恢复。
Objective:To investigate the effect of eleetroacupuncture combined with bone marrow mesenehymal stem cells (BMSCs)transplantation on inflammatory response and expression levels of neurotrophic factors in injured tissues of rats with spinal cord injury (SCI).Methods:50healthy adult SD rats were randomly divided into normal group,model group,eleetroacupuncture group,BMSCs group and combined group,10 rats in each group.Except for the normal group and the model group, the other groups received eleetroacupuncture,BMSCs or combined treatment at 1 week after SCI,and the intervention lasted for 12weeks.Limb movement function of rats in each group was evaluated by BBB score at the fourth,eighth and twelfth week after treatment,and the expression levels of interleukin-6(IL-6),tumor necrosis factor α(TNF-α)and interleukin-1β(IL-1β) proteins in injured tissues of spinal cord were quantitatively deteoted by Western-blot.The expression levels of brain derived neurotrophic factor (BDNF)and nerve growth factor (NGF)were determined by immunohistoehemical method at the .fourth and twelfth week after treatment.Results:Compared with the normal group,BBB scores of the other groups at fourth,eighth and twelfth week after treatment, the expression levels of BDNF and NGF at the fourth and twelfth week after treatment were significantly lower (P <0.05).BBB scores of the electroacupuncture group,the BMSCs group and the combined .group at the fourth, eighth and twelfth week after treatment,and the expression levels of BDNF and NGF at the fourth and twelfth week after treatment were significantly higher than those in the model group (P <0,05).BBB scores,expression levels of BDNF and NGF in the combined group at different time points were significantly higher than those in the electroacupuncture group and BMSCs group (P <0.05).The expression levels of IL-6,TNF-α and IL-1β in the electroacupuncture group,BMSCs group and the combined group were significantly lower than those in the model group at the fourth and eighth week after treatment (P <0.05).Conclusion:Electroacupuncture combined with BMSCs transplantation may inhibit the inflammatory response in rats with SCI,and increase neurotrophic factors and nerve growth factors,so as to promote the.recovery of motor function in rats.
