摘要
目的:研究甲胎蛋白(alpha-fetoproteinAFP)对人肝癌细胞信息传递物质Ca2+浓度的影响。方法:采用由脐带血血清中提纯的AFP作用于体外培养的人肝癌Bel7402细胞,通过共聚焦显微镜扫描,观察不同时间细胞内Ca2+浓度的变化。结果:在AFP作用4min时能显著提高Bel7402细胞内Ca2+浓度,与对照组比较升高204.1%,随后缓慢下降,到作用10min时降到正常水平。结论:AFP能通过改变细胞Ca2+浓度从而调节人肝癌Bel7402细胞内信息传递。
Objective: To investigate the effect of alpha-fetoprotein (AFP) on concentration change of Ca2+ in the Bel7402 cells of human hepatoma. Method:Purified AFP from human cord blood serum was used to act on the hepatoma Bel7402 cells culture in vitro, and the concentration changes of Ca2+ were observed by confocal microscopic scanning. Results: It showed that Ca2+ concentration of the cells was significantly increased 4 minutes after treatment, which enhanced 204.1% compared with the control group, afterwards it was slowly decreased and reached the normal level 10 minutes after treatment. Conclusion: AFP might regulate the messenger transduction of the Bel7402 cells by means of changing the concentration of intracellular Ca2+ .
