摘要
目的研究犬小肠缺血再灌注后 C-fos,PCNA 和 Bax 的表达及其意义.方法通过阻断分布范围较小的细小小肠动脉,建立缺血再灌注模型,以免疫组织化学方法研究小肠缺血再灌注0,30,60min和对照组动物小肠组织的 C-fos,增殖性细胞核抗原(proliferating cell nuclear entigen,PCNA)和 Bax 的表达特点.结果小肠缺血再灌注后 C-fos,PCNA 和 Bax 在粘膜层、粘膜下层和肌层的表达均有明显增加.在再灌注30min,三者表达达高峰,但至60min 则有明显下降.C-fos 在粘膜单层柱状上皮的阳性细胞率于再灌注0,30,60min 和对照组动物分别是89%,95%,53.4%和80%,在小肠腺分别为45%,90%,56%和81%,小肠粘膜上皮的 PCNA 阳性细胞率分別为95%,95%,55%和80%,在小肠腺则为82%,95%,60%和80%,但Bax 在小肠粘膜上皮的阳性细胞率分别是60%,95%,60%和10%,而在小肠腺则为30%,80%,10%和10%.结论小肠缺血再灌注后 C-fos,PCNA 和 Bax 的表达明显增加,说明它们的活动与小肠的缺血再灌注后细胞凋亡、增殖、恢复重建及调节功能有关.小肠小范围的缺血再灌注后 C-fos,PCNA 和 Bax 的表达具有发展快,但恢复也均较迅速的特点.本研究结果提示,在小肠手术中,选择较小的动脉进行阻断并缩短其时间可能对减少再灌注损伤和改善伤口修复有益.
AIM To investigate the expression and the implication of C- fos,PCNA and Bax on the intestinal ischemia and reperfusion injury in dogs. METHODS The intestinal ischemia and reperfusion model was established by clamping the intestinal artery distributing in small area,and the expression of C-fos,proliferating cell nuclear antigen(PCNA)and Bax were studied in intestinal tissue after ischemia and reperfusion at 0,30 and 60 min and control amimals by immunohistochemical method. RESULTS The expressions of C-fos,PCNA and Bax in mucosa,submucosa and muscularis increased obviously compared with that of normal animal,which reached a peak at 30 min and decreased at 60 min after reperfusion.The positive cell rate of C-fos after reperfusion for 0,30 and 60 min and normal animals was 89%,95%,53% and 80% in the simple columnar epithelium;and 45%,90%,56% and 81% in intestinal gland.For PCNA,the positive cell rate in simple columnar epithelium was 95%,95%,55% and 80%;82%, 95%,60% and 80% in intestinal gland,however,that of Bax was 60%,95%,60% and 10% in simple columnar epithelium; and 30%,80%,10% and 10% in intestinal gland respectively. CONCLUSION The expressions of C-fos,PCNA and Bax in intestinal ischemia and reperfusion are increased significantly, which demonstrated that C-fos,PCNA and Bax are related to the apoptosis,proliferation,recovery,reconstruction and regulating function in ischemia and reperfusion injury.The changes of C-fos,PCNA and Bax in ischemia and reperfusion on the limited area of intestine had the characteristics of quick development and restoration.It might play an important role in attenuating injury and improving the wound healing if clamping smaller arteries and shortening the time in intestinal operation.
