苦参碱诱导人肝癌细胞分化、凋亡时对G_1细胞周期调节因子的调控 被引量:85

摘要 目的:观察苦参碱诱导人肝癌细胞分化、凋亡时,G1细胞周期调节因子的变化,探讨苦参碱对肿瘤细胞增殖的调控。方法:0、0.3、0.8、1.5g/L苦参碱作用HepG2细胞3天,免疫组化检测p53、Rb、p21、p27、p16、cyclinD1蛋白表达;原位杂交检测p53、cyclinD1mRNA的表达。真彩色图像分析对基因表达强弱进行定量,结果用MicrosoftExcel软件统计处理。结果:用药后细胞周期负调控因子p53、Rb、p21、p27、p16表达增强;正调控因子cyclinD1表达减弱。结论:苦参碱诱导人肝癌细胞HepG2分化、凋亡的机制可能与苦参碱上调了G1细胞周期负调节因子的表达,下调了G1期正调节因子cyclinD1表达有关。 Objective: To observe changes of cell cycle G1 phase regulation factors during matrine induced differentiation and apoptosis in hepatoma cells, and to investigate the function of matrine in regulating proliferation of tumor cell. Method: HepG2 cells was treated for 3 days with 0, 0.3, 0.8, 1.5 g/L matrine. The protein expression of p53,p21,p27,p16,Rb,cyclin D1 was detected by immunohistochemistry and mRNA expression of p53,cyclin D1 was determined with in situ hybridization stain method. All results were analyzed quantitatively with high fidelity colored picture and statistically with Microsoft Excel Soffware. Result: After matrine treatment, the expression of cell cycle negative regulation factors p53, p21, p27, Rb, p16 were increased; while the expression of positive regulation factor cyclin D1 was decreased. Conclusion: The mechanism of matrine induced differentiation and apoptosis in HepG2 may relate to that matrine upregulate the expression of G1 cell cycle negative regulation factors and downregulate the expression of G1 cell cycle positive regulation factor cyclin D1.
