摘要
目的 :该研究旨在采用分子生物学技术 ,从基因水平上对深圳市某医院及河北辛集地区发生术后及注射后龟分枝杆菌暴发感染的临床分离株进行鉴定 ,并建立一套快速的的鉴定龟分枝杆菌脓肿亚种的探针杂交方法。方法 :根据龟分枝杆菌脓肿亚种标准株 16S - 2 3SrDNA间隔区序列测序结果 ,设计一对寡核苷酸探针 ,对其敏感性、特异性及其对临床标本和临床分离株的检测进行了研究。结果 :探针检测PCR扩增产物敏感性为 1ng ,只与龟分枝杆菌脓肿亚种杂交 ,与 5 6株龟分枝杆菌脓肿亚种临床分离株全部杂交 ;检测 5 7株临床标本阳性率为 6 6 .6 %。结论 :探针检测从基因水平上再次确认引起这两次暴发感染的致病菌为龟分枝杆菌脓肿亚种 ,结果快速可靠。
To determine the pathogen of the infection outbreak in Shen Zhen and He bei at gene level with molecular biological technique,and to establish a rapid identification method of M.chelonae subsp.abscessus by dot blot.We designed a oligonucleitide probe and the sensitivity and specificity of the probe were studied.The results showed:the sensitivity of the probe detecting PCR products was 1ng.Only M.chelonae subsp.abscessus was hybridization with the probe,the positive rate of the probe detecting clinical specimen was 66.6%.
