桦褐孔菌多糖对抗痨药致小鼠肝损伤的保护作用研究 被引量:5

摘要 目的:探讨桦褐孔菌多糖对异烟肼和利福平合用致小鼠肝损伤的保护作用。方法:48只小鼠随机分为对照组、模型组、护肝片组和桦褐孔菌多糖低、中、高剂量组各8只,除对照组外,其余各组每天采用异烟肼和利福平灌胃造成肝损伤模型;灌胃2 h后对照组和模型组分别予以等体积生理盐水灌胃,其余各组分别予以护肝片、桦褐孔菌多糖灌胃,连续两周。肝组织病理学检查,检测肝组织匀浆丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性。结果:肝组织病理学检查,桦褐孔菌多糖组明显减轻肝细胞变性和坏死的程度,缓解肝组织的病理改变。桦褐孔菌多糖组可降低肝匀浆MDA的含量,并能显著提高肝匀浆SOD的活性。结论:桦褐孔菌多糖对异烟肼和利福平致小鼠肝损伤具有明显的保护作用,其作用机制可能与其抗氧化作用有关。 Objective: To explore the protective effect of Inonotus obliquus polysaccharide (IOP) on hepatic injury in mice in- duced by isoniazid (INH) combined with Rifampicin (RFP) . Methods: Forty eight mice were randomly divided into control group, mod- el group, hepatic protection pill group, low Inonotus obliquus polysaccharide group, middle Inonotus obliquus polysaccharide group and high Inonotus obliquus polysaccharide group 8 mice in each group. Except the mice in control group, the mice in the other groups were treated with intragastric administration of isoniazid combined with rifampicin every day to establish hepatic injury model ; after two hours, the mice in control group and model group were treated with intragastric administration of normal saline of the same volume, the mice in the other groups were treated with intragastric administration of hepatic protection pills and Inonotus obliquus polysaccharide respectively for successive two weeks ; hepatic pathological examination was conducted, the malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were detect in liver hemogenate. Results : Histopathological examination showed that Inonotus obliquus polysaccharide could alleviate the de- generation and necrosis of hepatic cells and the pathological change of hepatic tissue. Inonotus obliquus polysaccharide reduced MDA content in liver homogenate and increased the of SOD activity in liver homogenate. Conclusion: Inonotus obliquus polysaccharide has obvious protective effect on hepatic injury in mice induced by isoniazid combined with rifampicin, which may be related to its antioxidation.
