摘要
目的探讨雄激素受体(androgen receptor,AR)在不同级别脑胶质瘤的表达及意义。方法收集12例正常脑组织和73例不同级别脑胶质瘤组织标本,其中WHOⅠ级8例,Ⅱ级15例,Ⅲ级23例,Ⅳ27例。采用免疫组织化学染色法和实时荧光定量PCR检测AR蛋白及AR mRNA表达情况,并分析正常脑组织和不同级别胶质瘤间AR表达差异。结果各级别胶质瘤组织中的AR蛋白阳性率和mRNA水平均明显高于正常脑组织(均P<0.05)。胶质瘤WHO分级与AR阳性细胞率呈显著正相关(rs=0.584,P<0.001),胶质瘤WHO分级与AR mRNA表达呈显著正相关(rs=0.885,P<0.001)。结论 AR在胶质瘤组织中表达增加,其表达水平与病理级别密切相关,提示AR可能在脑胶质瘤的生物学行为中发挥重要作用。
Objective To investigate the expression and its significance of androgen receptor (AR) in different grade human gliomas. Methods Seventy-third glioma tumor,samples and 12 normal brain samples were collected, including WHO grade Ⅰ in 8 patients, grade Ⅱ in 15, grade Ⅲ in 23 and grade Ⅳ in 27. The expression of AR protein was detected by immunohistochemistry, and AR mRNA by quantitative real-time PCR. The differences in the expression of AR protein and mRNA between normal brain and different grade gliomas were analyzed. Results The positivity rate of AR protein and expressiofi of AR mRNA in glioma tissues of different grades were significantly higher than those in normal brain tissues (all P 〈 0.05). The WHO classification of glioma was positively related to rate of AR-positive cells (rs = 0.584, P 〈 0.001). The WHO classification of glioma was also positively related to expression of AR mRNA (rs=0.885, P 〈 0.001). Conclusions The expression of AR increases in glioma and the expression level of AR is related to the pathological grades of glioma, which suggests that AR may play a critical role in the biological behaviors ofgliomas.
