阳和平喘颗粒对哮喘大鼠的保护作用 被引量:6

Experimental study of protection and its mechanism of Yanghepingchuan granules on asthmatic rats

摘要 目的:研究阳和平喘颗粒对哮喘模型大鼠的保护作用及其可能的作用机制。方法:将60雄性只大鼠随机分为正常对照组、模型组、阳和平喘颗粒(25.92、12.96、6.48g/kg)组、桂龙咳喘宁胶囊(1.62g/kg)组。试验开始第一天,大鼠腹腔注射10%卵清蛋白(OVA)氢氧化铝生理盐水溶液致敏,第8天再次致敏,正常对照组注射等量生理盐水。于初次致敏第15天起,除正常组外,其余各组大鼠用含1%OVA的生理盐水超声雾化激发20min,隔天一次,共7次,复制大鼠哮喘模型。实验第15天起,给药组分别灌胃给予相应的药物,治疗2周,正常组和模型组给予等容积溶媒。实验结束后,分别采用硝酸还原酶法和放射免疫法测定血清中一氧化氮(NO)、内皮素-1(ET-1)的含量,HE染色法观察哮喘大鼠肺组织病理改变,免疫组化法观察诱导型一氧化氮合酶(iNOS)在肺组织中的表达。结果:与正常组比较,模型组大鼠出现明显的哮喘症状,肺组织及周围有大量炎性细胞浸润,血清中NO、ET-1含量显著升高,肺组织中iNOS的表达水平明显高于正常对照组;与模型组相比,阳和平喘颗粒(25.92、12.96g/kg)组哮喘大鼠哮喘症状及肺组织炎症反应明显减轻,大鼠血清中NO、ET-1的含量及肺组织iNOS活性显著降低。结论:阳和平喘颗粒对OVA介导的大鼠哮喘模型的保护作用可能与其对ET-1、NO、iNOS的调节有关。 Objective: To observe the protection of Yanghepingchuan( YHPC) Granules and its possible mechanism of actions in asthmatic rats model. Method: 60 male SD rats were randomly divided into six groups,Control,Model,YHPC Granules( 25. 92,12. 96,6. 48g /kg),Guilongkechuanning( 1. 62 g /kg) groups. On the first day of the test,the rats were sensitized by intra-peritoneal injection of 10% ovalbumin( OVA) saline solution and sensitization once again on the eighth day.15 days later excitation,in addition to the Control group,the rest rats were atomized with 1% ovalbumin once every two days and 7 times. 15 days after sensitization,corresponding drugs were given for 14 days the model group and control group given distilled water. After treating,the level of NO and ET-1were detected in rats serun by nitrate reductase method and radioimunoassay,and the level of the activity of iNOS were detectes in lung was detected by enzyme linked immune method. Pathological change of lung tissue was observed by HE staining. Result: Compared with control group,in model group rats appear obvious symptoms of asthma,lunge tissue and surrounded were infiltrated by a large number of inflammatory cells. the levels of NO and ET-1 in serum was significantly increased( P < 0. 01),and the activity of iNOS in lung was relived significantly( P < 0. 01). Compared with model group,the YHPC granules( 25. 92,12. 96g /kg) could reduce asthma symptoms and the inflammatory cell infiltration of lung was relived,the levels of NO,ET-1 and the activity of iNOS in serum and lung tissue were decreased significantly( P < 0. 05,P < 0. 01). Conclusion: The YHPC granules could play anti-asthmatic effects and its functional mechnism may be related to the regulation of NO,ET-1 and iNOS.
