摘要
【目的】观察六味补气胶囊对慢性阻塞性肺病(COPD)大鼠T细胞介导转录调节因子(T-bet)、GATA-结合蛋白3(GATA-3)、孤独核受体(RORgammat)的影响。【方法】将50只SD大鼠随机分为正常组,模型组,六味补气高、中、低剂量组(剂量分别为0.7、0.35、0.175 g·kg-1·d-1),金水宝组(剂量为0.495 g·kg-1·d-1),脾氨肽组(剂量为0.33 mg·kg-1·d-1)。除正常组外,其他组大鼠均采用烟熏加脂多糖(LPS)气管滴入方法复制COPD模型,造模周期28 d后给药,连续给药30 d。采用动物肺功能仪检测大鼠肺功能,酶联免疫吸附法观察大鼠血清干扰素γ(IFN-γ)、白细胞介素(IL)-4、IL-17表达,Western blot法检测肺组织T-bet、GATA-3、RORgammat蛋白表达。【结果】与正常组比较,模型组大鼠肺功能值降低,血清IL-4和肺组织GATA-3表达降低,IFN-γ、IL-17、Th1/Th2升高,肺组织T-bet、T-bet/GATA-3、RORgammat表达升高(P<0.05或P<0.01)。与模型组比较,六味补气组肺功能值升高,IFN-γ、IL-17、Th1/Th2、T-bet、RORgammat表达降低,GATA-3表达升高(P<0.05或P<0.01)。【结论】六味补气胶囊通过上调GATA-3表达和下调T-bet、RORgammat表达而改善肺功能。
Objective To observe the effect of Liuwei Buqi Capsules(LBC) on T box expressed in T cells(Tbet), GATA binding protein 3(GATA-3), retinoid-related orphan receptor gammat(RORgammat) in rats with chronic obstructive pulmonary disease(COPD). Methods Fifty rats were randomly divided into normal group, model group, low-, middle- and high-dosage LBC groups(in the dosage of 0.7, 0.35, 0.175 g·kg-1·d-1, respectively), Jinshuibao group(in the dosage of 0.495 g·kg-1·d-1), and spleen aminopeptidase group(in the dosage of 0.33 mg·kg-1·d-1). Except for the normal group, rats in the other groups were given smoking combined with transtracheal dripping of lipopolysaccharide(LPS) for 28 days to induce COPD. After modeling, the medication groups were given the corresponding medicine for 30 continuous days. Pulmonary function was detected by animal spirometer, and serum interferon gamma(IFN-γ), interleukin(IL)-4, and IL-17 were detected by enzymelinked immunosorbent assay( ELISA). Expressions of T-bet, GATA-3, RORgammat protein in lung tissue were detected by Western blotting. Results Compared with the normal group, pulmonary function values, serum IL-4 level and the pulmonary GATA-3 protein were decreased, and serum levels of IFN-γ, IL-17, Th1/Th2 as well as the expression levels of T-bet, T-bet/GATA-3, and RORgammat protein in the lung tissue were increased in the model group(P〈0.05 or P0.01). Compared with the model group, pulmonary function was improved, serum levels of IFN-γ, IL-17, and Th1/Th2, as well as T-bet and RORgammat protein expression levels were decreased, and GATA3 protein expression was increased( P〈0.05 or P0.01). Conclusion LBC shows an effect on improving pulmonary function by up-regulating GATA-3 expression and by down-regulating T-bet and RORgammat expression.
