摘要
目的观察补阳还五汤对全脑缺血再灌注后大鼠皮层神经元NR1 mRNA表达的影响,研究补阳还五汤抗全脑缺血后再灌注损伤的分子机制。方法 SD大鼠随机分为3组,即正常组、模型组和药物组(补阳还五汤预干预),模型组和药物组再分别设置2,24h两个观察时间点。连续给药5 d后,采用改良的Pulsinelli四血管闭塞法复制全脑缺血大鼠模型,再灌后2,24 h采用RT-PCR技术检测各组大鼠皮质神经细胞NMDA受体(N-methyl-D-aspartic acid receptor)亚单位NR1的mRNA表达水平。结果与正常组比较,模型组2h,24h等2个不同时间点,NR1的mRNA表达水平显著升高(P<0.05);与模型组比较,药物组再灌注后2h,24h等2个不同时间点,NR1 mRNA表达水平显著降低(P<0.01)。结论补阳还五汤预干预对全脑缺血再灌注模型大鼠大脑皮层神经元NR1 mRNA表达水平的增高有调低作用。
Objective To study the molecular mechanism of Buyang Huanwu Decoction ( BYHWD ) , a famous prescription for supplementing Qi and activating blood circulation recorded in Corrections of Errors in Medical Works written by Ye Tianshi in Qing dynasty, in preventing the global cerebral ischemia - reperfusion injury. Methods A total of 66 Sprague - Dawley rats were randomly divided into normal group, model group and BYHWD pretreated group. Rats in the model group and the BYHWD pre- treated group were subdivided into 5 groups, according to 2 - and 24 - hour reperfusion. Five days after the administration of BY- HWD, rats in the model group and the BYHWD pretreated group were induced into global ischemia - reperfusion model using Pulsinelli four - vascular - occlusion method. The expression levels of the subunits NR1 mRNA of the N - methyl - D - aspartic acid receptor (NMDAR) in the rat cortex of all groups were determined after 2 -and 24 -hour reperfusion, respectively. Com- pared with those in the normal group, the expression levels of NR1 mRNA in the cortical neurons of rats in the model group were significantly increased at all intervals after the reperfusion (P 〈 0.05). Compared with those in the model group, the expression levels of NR1 mRNA in the BYHWD pretreated group were significantly decreased at all intervals after the reperfusion ( P 〈 0. 05). Results Conclusion BYHWD can prevent the increasing of NR1 mRNA expression levels in rats with isehemia - reperfu-sion injury.
