摘要
目的探讨200 mmol/L高渗盐液(HS)对严重烫伤大鼠肠组织的氧化应激以及外周血单个核细胞(PBMCs)中的p38丝裂原活化蛋白激酶(p38MAPK)活性的影响。方法按照随机抽样法将SD大鼠划分为3组:对照组(8只)、乳酸钠林格液组(LR组,24只)以及高渗盐液组(HS组,24只)。烫伤前48 h 3组大鼠均予以颈静脉置管,对照组的大鼠直接处死取材。LR组和HS组的大鼠在经过30%体表面积烫伤(TBSA)后分别予补充LR与HS进行复苏治疗,并分别于烫伤补液后2、8、24 h处死取材。检测3组大鼠肠组织中髓过氧化物酶(MPO)的活性与丙二醛(MDA)的含量,分离各组的PBMCs并采用Western blot法检测细胞中p38MAPK的活性,将所得数据进行统计学分析。结果与对照组相比,HS组及LR组肠组织中MPO活性和MDA含量明显增高,差异有统计学意义(P<0.05,P<0.01),HS组与LR组相比伤后各时间点上肠组织MPO活性与MDA含量明显降低,差异有统计学意义(P<0.05,P<0.01),而HS组和对照组相比伤后3个时间点上肠组织MPO活性与MDA含量无明显区别,差异无统计学意义(P>0.05)。HS组与LR组复苏后各时间点PBMCs中的p38MAPK活性表达与对照组比较显著增强,差异有统计学意义(P<0.01),但LR组和HS组进行比较,差异无统计学意义(P>0.05)。结论使用200 mmol/L的HS进行液体复苏较LR能明显减轻严重烫伤大鼠肠组织的氧化应激反应,两组对PBMCs中的p38MAPK活性表达无差异。
Abstract Objective To study the effect of 200 mmol/L Na^+ hypertonic saline resuscitation on oxidative stress of intestinal tissue and p38MAPK expression of peripheral blood mononuclear cells (PBMCs) in severely burned rats. Methods Fifty-six SD rats were randomly divided into three groups: control group, sodium lactate Ringer's solu- tion group (LR group) and hypertonic sodium solution group (HS group). Neck vein catheterization was used on all rats, and burn resuscitation therapy was adopted in the rats of LR group and HS group. The myeloperoxidase (MPO) activity and malondialdehyde (MDA) content of intestinal tissue were measured in three groups. PBMCs were isolated and the p38MAPK expression was determined by Western blot analysis. The data drawn from the test were statistically analyzed. Results The MPO activity and MDA content of intestinal tissue at each time point after resuscitation between HS group and LR group were significantly higher than those of the control group ( P 〈 0.05 , P 〈 0. 01 ). The MPO activity and MDA content of intestinal tissue at each time point after resuscitation in HS group were significantly lower than those of the LR group (P 〈 0. 05, P 〈 0. 01 ), which compared with the control group, the difference was not statistically significant (P 〉 0.05 ). The p38MAPK expressions of PBMCs at 2, 8, 24 h after fluid resuscitation between LR group and HS group were significantly expressed in comparison with the con- trol group (P 〈0. 01 ). They had no obviously difference between LR group and HS group (P 〉 0. 05 ). Conclu- sion Compared with sodium lactate Ringer's solution, 200 mmol/L Na^+ hypertonic saline resuscitation can attenu- ate oxidative stress of intestinal tissue in severely burned rats, but not affect the p38MAPK expression of PBMCs in severely burned rats.
