摘要
以叶黄素为受试物,采用抗氧化能力评价化学方法,包括氧自由基吸收能力法(ORAC方法)、Trolox Equivalent Antioxidant Capacity Assay(TEAC法)、Folin-Ciocalteu法(FCR实验法)、1,1-二苯基-2-三硝基苯肼法(DPPH法),同时采用评价生物大分子损伤的脱细胞彗星实验,对叶黄素的抗氧化-促氧化作用进行探索。结果表明,ORAC法和DPPH法表明不同浓度叶黄素分别呈现出抗氧化和促氧化的双重作用;TEAC法表明叶黄素具有抗氧化作用、FCR实验表明叶黄素含有酚类物质较少,酚类物质不是叶黄素抗氧化作用的主要因素;脱细胞彗星实验表明高剂量叶黄素对DNA分子具有损伤。因此,叶黄素在不同浓度表现出抗氧化—促氧化的双重作用。
In the study, lutein was used as the test material, chemical methods including Oxygen Radical Absorbance Capacity(ORAC) ,Trolox Equivalent Anti-oxidant Capacity assay(TEAC), Folin-Ciocalteu Reaction ( FCR), 1,1- Dfphenyl-2- picrylhydrazylradical2,2- Diphenyl- 1 - (2,4,6-trinitrophenyl ) hydrazyl ( DPPH ) were used to evaluate the anti-oxidative character of lutein, and at the same time, the method of evaluating biological macromolecular injury-acellular comet assay was applied in this study.Different concentrations of lutein was demonstrated to have anti-oxidation and pro-oxidation in the ORAC and DPPH;lutein was proved to have anti- oxidation in TEAC;lutein was indicated to contain less phenolics,and the phenolics was not the main factor of anti -oxidant effect in FCR;high concentration of lutein had damage to DNA in the acellular comet assay.Therefore, lutein showed double effect of anti-oxidant and pro-oxidant in different concentration.
