百合地黄汤对创伤后应激障碍大鼠行为学及海马GR/MR表达的影响 被引量:20

Effects of Baihe Dihuang Decoction on Behavior and GR /MR Expression in Hippocampus of Rats with Post Traumatic Stress Disorder

摘要 目的:通过观察百合地黄汤对创伤后应激障碍(PTSD)大鼠行为学改变及海马GR/MR表达的影响,探讨百合地黄汤对PTSD的干预作用。方法:将60只SPF级雄性SD大鼠,随机分为正常组、模型组、阳性对照组、低剂量组、中剂量组和高剂量组,每组10只。正常组给予常规无干扰喂养2周,余5组采用SPS法造模后,低剂量组、中剂量组和高剂量组给予不同浓度百合地黄汤煎液,阳性对照组予氟西汀,模型组予等量生理盐水。2周后各组大鼠采用旷场实验(OF)进行行为学评定,并用免疫印迹法检测大鼠海马GR/MR水平。结果:模型组大鼠各行为学指标及GR/MR表达水平与正常组均存在显著性差异(P<0.01);中、高剂量百合地黄汤及氟西汀可增加模型大鼠的水平活动距离和直立、修饰次数,降低呆滞次数和排便量;尤其是高剂量百合地黄汤与氟西汀相比能显著增加直立(P<0.05)、修饰(P<0.01)行为;不同剂量百合地黄汤及氟西汀均能显著下调海马GR表达,上调MR表达,且各组无明显差别。结论:调节海马GR/MR的表达可能是百合地黄汤对PTSD大鼠的干预治疗机制之一。 Objective:To observe the effects of Baihe dihuang decoction on behavior changes and GR/MR expression in the hippocampus of PTSD rats and to explore the mechanism of therapeutic actions.Methods:60 rats were randomly divided into normal group,model group,positive group,low,medium and high dose Baihe dihuang decoction groups.The three groups were given different concentration decoction respectively,positive group was given fluoxetine hydrochloride,and SPS model group was administered with equal volume of normal saline.The length of administration period was 2 weeks.Each rat was detected by open field text and executed to examine GR/MR expression in hippocampus by Western blot.Results:Three doses of Dihuang baihe decoction groups and positive control group were able to obviously increase horizontal movement distances,upright behavior,modification behavior and decrease sluggish behavior,stool quantity.Furthermore,high doses of Baihe dihuang decoction could significantly increase upright behavior(P < 0.05)and modification behavior(P < 0.01)to compare with positive control group.Three dose of Dihueng baihe decoction groups and positive control group were able to remarkably influence on GR/MR expression in hippocampus to compare with model group (P <0.01),though comparison among medicine groups were not statistically significant(P > 0.05).Conclusions:Baihe dihuang decoction do have therapeutic effects on PTSD rats,and the influence on GR/MR expression in hippocampus is one of the possible mechanisms.
