摘要
以对硝基β-D-葡萄糖苷为底物,对茶叶中β-D-葡萄糖苷酶活性测定条件进行了研究,包括缓冲液pH值、底物浓度、反应温度和时间等条件对酶活性的影响。结果表明,20mg茶树鲜叶粗酶提取液底物浓度为10mmol/L,在49℃条件下反应120min能得到最大的光吸收值。
The conditions including substrate concentration,buffer pH,reaction
temperature and time etc.for testing -D-glucosidase activity in fresh tea leaves using
p-Nitrophanyl -D-Gluopyranoside (pNPG) as substrate were studied.The results showed that 20
mg enzyme extracts from fresh tea leaves gave the highest absorbance when reacted with
PNPG at the concentration of 10 mmol/L under the conditions of buffer pH60,reaction
temperature 49 and reaction time 120 minutes.
