水稻无性系变异的2种重复DNA甲基化分析

摘要 对14份发生分离性状的非整倍体无性系进行染色体水平的检测,发现原始系和变异株有丝分裂过程正常,未发生染色体水平变异。对其进行DNA甲基化分析发现不管是原始系与变异株之间,还是不同年份培养的无性系之间在CentO和Os48这2种重复序列甲基化水平没有明显的差异。此外,在染色体水平也没有发现明显全基因组甲基化的差异。 ABSTRACT: During asexual reproduction, cells divide by mitosis, so the genotype of the daughter cells should be the same as that of the original parent cell. However, somaelonal variation can occur sometimes during the process of tissue culture. In this study, among the 14 somaelonal variation lines without chromosome number variation, no abnormal mitotic chromosome behavior was detected. The DNA methylation analysis showed that they had no obvious differences in DNA methylation for the two major tandem repeats CentO and Os48 from centromerie regions and telomeric regions, respectively. In addition, no obvious methylation change was detected in whole chromosome level.
