摘要
为了探明山杏扦插繁殖生根机理,并为山杏良种的无性快繁提供理论依据,以山杏嫩枝为插穗,用IBA、NAA和IBA+NAA处理插穗,以清水处理为对照,研究扦插生根过程中插穗内源激素含量的变化。结果表明:山杏嫩枝扦插生根类型属于愈伤组织生根型,插穗生根过程可划分为诱导、表达和伸长3个阶段。在插穗生根过程中,植物生长调节剂处理能够引起插穗内源激素含量的变化,从而促进生根,IBA+NAA、IBA和NAA处理插穗的IAA含量在扦插后10 d时比对照分别提高53.3%、34.7%和25.8%,扦插后40 d时比对照分别提高65.7%、56.6%和28.2%。ABA、GA3含量在总体变化趋势中呈波动下降,在生根诱导阶段,ABA、GA3含量下降趋势尤为明显。ZT含量变化呈现先升高、后降低的变化趋势,峰值出现在诱导末期。ABA、GA3和ZT含量与生根率呈负相关,各处理的IAA含量、IAA/ABA值、IAA/ZT值在诱导和伸长阶段出现2次峰值,与对照差异显著,且均与生根率呈正相关。
In order to ascertain rooting mechanism in Armeniaca sibirica cutting propagation, and provide a theoretical basis for rapid asexual propagation of fine cultivars, the softwoods ofA. sibirica were taken as the cuttings which were treated by IBA, NAA, IBA + NAA and water, and variation of endogenous hormone contents in softwood cuttings was studied during adventitious root formation. The results show that the type of adventitious root formation in softwood cuttings of A. sibirica belonges to the callus type. The period of adventitious root formation can be divided into three stages, which include induction, expression and prolongation. During the formation of adventitious root, the plant growth regulator treatments can promote rooting through affecting endogenous hormone contents. On the 10th day after cutting, IAA content in the cuttings treated by IBA+NAA, IBA and NAA are higher by 58.3%, 37.4%and 25.8% than the control, and higher by 65.7%, 56.6% and 28.2% than the control on the 40th day after cutting. ABA and GA3 contents present downward trend in general changes. And at induction period, the downward trends of ABA and GA3 contents are more obvious. ZT content has the trend of increased firstly and then decreased, and the peak value occurs at late induction period of adventitious root formation. ABA, ZT and GA3 contents are negatively correlated with rooting rate. IAA content, ratio of IAA to ABA and ratio of IAA to ZT have two peaks at induction period and prolongation period of adventitious root formation, which are significantly different from the control and are positively correlated with rooting rate.
