铁皮石斛多糖的提取工艺优化及其抗氧化活性研究 被引量:13

Optimization of polysaccharides extraction from Dendrobium officinale Kimura et Migo and investigation on its antioxidant activity

摘要 【目的】优化铁皮石斛多糖的提取工艺,并评价铁皮石斛多糖的抗氧化活性。【方法】以铁皮石斛多糖提取率为响应值,在单因素试验基础上,以提取时间、提取次数及液(mL)料(g)比为试验因素,采用响应面法建立数学模型,筛选最佳提取工艺条件。采用自由基清除能力体系初步评价铁皮石斛多糖的抗氧化活性。【结果】通过二次回归模型响应面分析,获得最佳的铁皮石斛多糖提取工艺为:提取次数3次、提取时间2h、液(mL)料(g)比75。在此最佳工艺条件下,铁皮石斛多糖提取率为34.96%,与理论值(36.57%)相对误差小于5%。铁皮石斛多糖对DPPH和ABTS自由基的半数清除率分别为1.20和3.65mg/mL。【结论】采用响应面法优化得到了铁皮石斛多糖的最佳提取工艺,该工艺方便可行,得到的多糖具有较强的抗氧化活性。 【Objective】This study aimed to optimize polysaccharides extraction from Dendrobium officinale Kimura et Migo and investigate its antioxidant activity.【Method】The response surface method (RSM) based mathematical model was established using polysaccharides extraction rate as response value,and using extraction time,extraction frequency,and ratio of water to feedstock as experimental factors to screen optimum extraction conditions for polysaccharide extraction from D.officinale Kimura et Migo.Antioxidant activity of polysaccharides was studied by DPPH and ABTS free radical elimination method.【Result】The optimum conditions obtained by RSM were:number of extraction times was 3,duration of extraction was 2 h and ratio of water to feedstock was 75 mL/g.Under the optimal conditions,the predicted extraction rate was 36.57%,while the experimental extraction rate was 34.96%, with a difference of less than 5%.IC50 of DPPH and IC50 of ABTS were 1.20 and 3.65 mg/mL,respectively.【Conclusion】Optimization of the polysaccharide extraction from D.officinale Kimura et Migo by RSM was convenient and feasible, and obtained polysaccharides had good antioxidant activity.
