摘要
目的:采用石墨炉原子吸收光谱法测定SD大鼠血清中铜的含量.方法:大鼠血清样品采用湿法消化前处理,灰化温度850℃,原子化温度2100℃条件下检测,结果:铜的线性范围为1.00~10.00 μg.L-1,r=0.9976,检出限为0.0850μg·L-1;加样回收率在99.1%~105.8%之间.日内、日间精密度RSD<5.0%(n=5);结论:所建立的石墨炉原子吸收法能够准确测定大鼠血清中铜的含量,方法具有重复性好,操作简便等特点,能为生物样品分析微量元素提供参考.
The copper concentrations in serum of rats are detected by graphite furnace atomic absorption spectrometry. The pyrolysis temperature and atomization temperature are optimized. The metllod to determine copper contents in serum of rats using wet digestion combined with graphite furnace atomic absorption spectrometry is established. The results show that the calibration curve is linear0=0.9976) within the range of 1.00-10.00 μg·L-1 for copper, the detection limit of this method is 0.0850 μg·L-1 the standard recovery rates are between 99.1% and 105.8%; the intra-day and inter-day precision RSD are less than 5% (n=5); The method is precise, accurate and suitable for analysis of trace biological samples. It can be used to provide feasible data on copper contents in animal phannacokinetic studies.
