摘要
目的探讨蛙血清去蛋白提取物对离体大鼠心肌缺血再灌注损伤(IRI)的保护作用。方法应用Langendorff恒压灌注大鼠心脏,停灌-复灌方式制备缺血再灌注模型。观察缺血再灌注期间左心室内压上升的最大变化速率(+dp/dt max)、左心室内压下降的最大变化速率(-dp/dt max)、左心室收缩压(LVSP)和冠状动脉流量(CF)、采用黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活性,硫代巴比妥酸法测定丙二醛(MDA)含量。结果蛙血清去蛋白提取物高、中浓度组能显著改善IRI引起的心功能损伤,升高LVSP、CF、+dp/dt max、和-dp/dt max,减少心肌组织中MDA的生成,增加心肌组织中SOD活性。蛙血清去蛋白提取物高浓度组比蛙血清去蛋白提取物中、低浓度组对上述指标的改善更为显著。结论蛙血清去蛋白提取物可改善IRI引起的心脏收缩及舒张功能障碍,增加心肌清除氧自由基的能力且这种保护作用存在量效关系。
Objective To investigate the protective effect of deproteinized frog serum extract on myocardium ischemia - reperfusion injury in isolated rat hears. Methods The method of hearts in vitro constant pressure perfusion was set up by Langendorff. The ischemia - reperfusion model was established by stopping and reperfusing to the hearts. The following indexes of rats in each group were measured re- spectively :left ventricular systolic pressure(LVSP) , maximal rate of the pressure ( + dp/dtmax ) increase, maximal rate of the pressure ( - dp/dtm^x) decrease,coronary flow( CF), the contents of superoxidc dismutase ( SOD), malondialdehyde (MDA). Results The group of deproteinized frog serum extract could improve cardiac function,increase the LVSP, CF, + dp/dt and -dp/dt reduce MDA and in- creased SOD in myocardial tissue. Compared with low and middle dose group, the protective effect of high dose group was more obvious. Conclusion The deproteinized frog serum extract can protect myocardium isehemia - reperfusion injury and lipid peroxidation,improve cardiac function and the protection has concentration -response relationship.
