摘要
为了研究核桃EST序列的通用性以及山核桃的遗传多样性,从NCBI数据库的核桃EST序列中设计了20对引物,并从中筛选出6对,对24份山核桃以及27份核桃样品进行分析,共检测到46个位点,其中多态性位点41个。利用Popgen32软件对6组引物进行分析。结果表明,观察等位基因数(N a)平均为1.869 6,有效等位基因数(N e)平均为1.623 4,基因多样性指数(H)平均为0.344 7,Shannon信息指数(I)平均为0.502 3。采用UPGMA法进行聚类分析,在相似系数为0.73时所有样品可明显区分为2个类群,说明由核桃EST筛选出的EST-SSR引物具有较好的通用性,可用于山核桃遗传分析、系统分类以及品种鉴定。
In order to study universality of EST sequences and genetic diversity of Carya cathayensis, 20 pairs of primers were designed from walnut EST sequences in the NCBI database, and then 6 pairs were selected out. 24 pecans samples and 27 walnut samples were analyzed. A total of 46 sites were detected, including 41 polymorphic sites. Six pairs of primers were analyzed by using Popgen32 software. The results show that the observed number of alleles (Na) is an average of 1.869 6, the effective number of alleles (No) is an average of 1.623 4, the Nei's gene diversity (H) is an average of 0.344 7, and Shannon information index (/) is an average of 0.502 3. The results of clustering analysis by using UPGMA method show that those samples can be divided into two groups when the similarity coefficient is 0.73. The EST-SSR primers selected have good universality, and can be used for C. cathayensis genetic analysis, classification and cultivar identification.
